AB STRACTDanshen films, biodegradable in coelom, were prepared for uses in promotion of surgical anastomosis in gastrointestinal tract.Danshen (Radix Salviae Miltiorrhizae), a traditional Chinese medine, which can promote healing of wound cuts, was used as a main ingredient in the film. A liquid extract of danshen active constituents, containing tanshinone 0.07390 mg/mi, was prepared, for which an ultrafiltration method was used. Assay of tanshinone was performed with L-IPLC method.Chitosan and gelatin ?the biocompatible and biodegradable natural polymers were selected for carriers in the film. Formulation design was carried out by test physicochemical properties including ability to form a film, tractility, flexility, intensity and uniformity of the film, and the biodegradability of the film in mice. The drug release and biodegradability in mice and in vitro of the film was studied. The results showed that the film was qualified for our essential requirements, and the film had significant effect to sustain the release of tanshinone II one of thebasic components of Salviae Miltiorrhizae.Pharmacodynamics of the film on surgical anastomosis in gastrointestinal tract was studied in rabbits, in which pathohistologic morph and expression of growthfactors in the cuts was observed. The film could improve the wound healing by regulating inflammatory reaction, collagen synthesis, and the expression of PDGF, TGF- ~, b-FGF, in recovery cells.and it is more effective than Salviae Miltiorrhizae injected.
