[Abstract] objective To establish an objective and quantitative criterion for the assessment of WT1 gene expression and prognosis of acute leukemia (AL) and minimal residual disease (MRD) in AL. Methods Expression of WT1 gene was measured with FQ-RT-PCR in 40 cases of AL at different disease-developing stage. Results Expression of WT1 was revealed in 26 of 40 AL patients and none of 12 normal controls. The CR rate (34.6%) was lower in WT1 positive group than that (78.5%, P<0.05) in WT1 negative group. The positive rates and the average levels of WT1 gene expression were significantly different among these 3 groups (69.2%,4.14×103copies/ml for untreated guoup;100% ,8.58×103copies/ml for replastic group and 33.3%,7.87×103copies/ml for CR>3 years). The WT1 gene expression level was markedly decreased after CR whereas highly increased when replased. Conclusion The method FQ-RT-PCR was objective and accurate. WT1 gene expression level was correlated with the treatment outcome and the prognosis of AL. WT1 gene is a new marker for the detection of MRD.
