AbstractDepartment of Oral Clinical Medicine, Medical College, Qingdao University Postgraduate Cai ZhenInstructor Fan Gongwei, Chen WantaoObjective To study the molecular basis of tumor cell translocation from the aspects of P-selectin mediating of tumor adhesion,in order to make known the secrets of malignant tumor cell metastasis, which can provide new ways block the metastasis of malignant tumor . Methods Using lipoprotein mediated DNA translocation the CHO cells were transfected with P-selectin, then selected by G418 and MTX and detected by Flow-type cell instrument, in order to obtain the highly -expressed P-CHO cells. By the Rolling Test ,we detected the rolling and binding of tongue squamous cell carcinoma cells with CHO cells which have P-selectin expression. Results Using the me- thod, we can obtain the highly expressed P-CHO cells. In the Rolling Test, the number of Tca8113 cells adhering to P-CHO com- pare with the number of Tca8113 cells adhering to CHO has statistical signi- ficance (P<0.001). After P-selectin adhesion-blockage monoclonal antibody and polyclonal antibody handling, the adherance of Tca 8113 cells with CHO has no changes (p>0.05). the number of Tca8113 ,s adherance to P-CHO after mAbG1 handling have statisti- cal significance (P<0.001), the number of adherance after PolyAb handling also have statistical significance (P<0.001), and the num- ber of Tca8113 cells adhering to P-CHO and CHO after mAbPS1 has no changes than before(p>0.05). Conclusion 1 Using cell culture study infiltrative growth and metastasis is better than using assay in vivo. 2 Flow cytometric assay is very important method to detect P-selectin Rg adhere CHO cells. 3Using lipoprotein mediated DNA translocation the CHO cells transfected with P-selectin, then select- ed by MTX and detected by Flow-type cell instrument, is a better way to obtain the highly -expressed P-CHO cells. 4.The adhering of Tca8113 cells to P-selecting has specificity and it has significant dep- endency on P-selectin. It can be concluded that P-selectin plan an important role in metastasis of human oral malignant tumor and it play a significant role in finding a good way to block the adhesion and metastasis of tumor. |
