Immunohistochemical Study on the Expression of TumorSuPpressor Gene Rb. p16 in Uygur and Han NasopharyngeaICarcinomas in XinjiangDepartment of OtorhinoIaryngology the First AffiliatedHospital of of Xinjiang Medical UniversityPostgraduatc: Qu Yaming Supervisor: Li TongyinglAbstractl Objective To study the expressions of Rb,p16protein in tumorous tissues of Uygur, Han patients withnasopharyngeal carcinomas(NPC) and their clinical significance.Methods The abnormal expressions of Rb' pl6 protein in 60NPC(Uygur 30,Han 30) and 37 non-tumor nasopharyngeal tissues(NP.Uygurl7,Han 20) in Xil1jiang were detected by the LSABimmunohistochemical method respectively. Results Thefrequencies of Rb, pl6 protein 30.0%, 4l .7% in NPC weresignificantly lower than NP 83.8%, 83.8% respectively(P<0.05).Significant differences existed in the expressions of Rb,p l6between NPC(23.3%,56.7%) and NP(76.5%,76.5%) inUygur(P<0.05).There were statistical differences in the frequenciesof Rb,pl6 protein between NPC(36.7%,26.7%) and NP(90%,90%) inHan. It was fOund that significant difference between UygurNPC(56.7%) and Han NPC(26.7%) in the frequency of pl6expression(P<0.05).The expressive rate of Rb protein was 83.3% inkeratinizing type, 6.7% in non-keratinizing type, l l. l%inundifferentiated type in Uygur NPC(P<0.05).NO correlationsbetween Rb,pl6 abnormal expressions and lymph node metastesis,clinical stage,surviva1 rate were fOund(P>0.05). Conclusions: l .Lowexpressions of pl6,Rb may involve the important effect onnasopharyngeal carcinogenesis of Uygur and Han in Xinjiang. 2.Theethnic genetic background may affect this carcinogenic progression.3.There is the correlation between Rh and histologic differentiation in Uygur NPC. 4.No correlations between Rb,p16 abnormal expression and lymph node metastesis. clinical stage. surrival time were foun&which implicate that the inactivaties ofpi6 and Rb protein may be early events in NPC pro~ress1on.
