Studies on Polysaccharide Extraction from Kelp and the Influence of It to Mouse Body Immunity (Shandong Teachers University Zhang Guixiang ) Abstract Kelp is a kind of large-scale alga and is planted w]dely in our country. Kelp is also a kind of foodstuff, which can he used as both medicine and food. In our country, kelp was eaten for a long period. In this thesis, polvsaccharide extraction and its influence to mouse body immunity based on the leftover bits and pieces of kelp were studied. First, qualitative and quantitative analysis of polysaccharide was set up. There are lots of measurements of polysaccharide .In this article, the oxidative developer silver nitrate was used for the qualitative analysis and sulfuric acid-phenol for the quantitative analysis. Then, extraction parameters were studied on kelp. It was found out that polysaccharide content of kelp in the extraction liquors was varied with extraction conditions, such as the pretreatment way, amount of infusion, soaking temperature and soaking time, etc. Two ways were used for pretreatment-steeping after high-temperature pretreatment and steeping without high-temperature pretreatment. It turned out that polysaccharide in the soaking liquids raised distinctly with the high-emperature pretreatment. Polysaccharide content of it was more than three times kelp without high-temperature pretreatment. li reaches to 30230 i-i g%g or also. Considering the amount of water, kelp with high? temperature pretreatment was used in the thesis as kelp to water equaling to 1:10, 1:20, 1:30, 1:40, 1:50. The result was that when kelp to water equals to 1:30, the pol\'saccharide in the soaking liquids was the highest. Along with temperature s ascending, the poivsaccharide in the soaking liquids is rising. But when the temperature surpasses centigrade 70, the polysaccharide content rises unclearly. As time goes on, the polysaccharide content rises. But when surpasses 8 hours, the influence by the time ~snt large. Little mice as subjects of experiment were used. These mice were inoculated S80 tumor. Then the mice were injected with soaking liquids. lt discovered that the tumor inhibition was affected by many factors, for example, soaking temperature, soaking time, equivalent concentration of soaking liquids. Along with temperature's ascending, the tumor inhibition of the soaking liquids increases. But when surpasses centigrade 70, the inhibition decreases. As time goes on, the tumor inhibition of the soaking liquids increases. But when surpasses 9 hours, it decreases. With the same soaking liquids, when concentrated to high concentration, the inhibition reduces, even increases the growth of tumor. When diluted to low concentration, the inhibition increases. Inthisarticle, preliminary separationwas carried. The soaking liquid was sediment by alcohol. When added to 30 per alcohol concentration, 60 per alcohol concentration and 80 per alcohol concentration, three kinds of po1 ysacchari de were obtained which were DT1, DT2 and DT3.
