Inducible Nitric Oxide Synthase and Vascular Endothelial Growth Factor Expression in Oral Squamous Carcinoma and Their Relation to Angiogenesis OBJECIVE To investigate inducible nitric oxide synthase (iNOS)and vascular endothelial growth factor (VEGF)expression in oral squamous carcinoma(OSC)and the relationship between the expression and the biological behaviors of OSC. METHODS Tissue sections from 58 OSC patients (38 men, 20 women, 35~58 years old) were examined immunohistochemically (SABC method) for protein expression of iNOS and VEGF. Micro-vessel density(MVD) was counted by endothelial cells immuno-stained by anti-CD34 antibody. RESULTS Positive iNOS immunostaining was detected in 32 cases, the positive rate was 55.2%, and the positive VEGF cases 36, the rate was 66.1%, the expressions of VEGF and iNOS were all positive were 31 cases, the rate was 53.5%.MVD was 35.13±5.35 /Hp in OSC positive for iNOS while it was 28.42±5.64/Hp in OSC negatively, t=4.59, P<0.01. The similar result was that MVD was 34.89±5.65/HP in VEGF positive group while it was 28.50±5.69/HP in VEGF negative group, t=4.09, P<0.01. The close relationship between the MVD and the mediate differentiation was found. The MVD of grade III and II was significantly denser than that in grade I, P<0.01, while the grade II group compared to grade group was P<0.05 and grade III group compared to grade I group was P<0.01. iNOS and VEGF positive expression rates were closely related to mediate differentiation either, the two positive rates in grade III and II compared with that in grade in grade I group were all P<0.05. The lymph node metastasis rate in iNOS and VEGF group was higher than the negative group, P<0.05, and the positive group's recurrent cases in 5 years were more than that of the negative group, P<0.05. CONCLUSIONS iNOS and VEGF high express in OSC, and they may play a important role in OSC growth, lymph node metastasis and recurrence; micro-vessel remarkably increase in OSC; iNOS and VEGF express closely related to tumor mediate differentiation, and they maybe increase tumor angiogenesis; iNOS, VEGF and MVD can be served as prognostic factors for OSC patients.
