ObjectiveTo study expression and significance of vascular endothelial growth factorand endothelial nitric oxide in endometriosis by immunohistochemistry.Material and Methods1 Clinical material: cases from the Kuangwujv General Hospital of FuShun betweenAugust 2004 and April 2007,including 30 cases of ovarian endometriosis;30 casesof normal endometrium。2 Immunohistochemistry was employed in this study to detect expression ofvascular endothelial growth factor (mouse anti-human VEGF monoclonalantibody;ready to use,Changdao Biotech)and endothe-lial nitric oxidesynthase(mouse anti-human eNOS monoclonal antibody;ready to use,ChangdaoBiotech).All procedures were implemented according to the productillustrations.We used PBS instead of primary antibody as negative control.The immunostaining of VEGF and eNOS was localized to the nucleus andcytoplasm.Statistical evaluation was performed using chi-square to differentiatethe rates of different groups.Results1. VEGF is expressed mainly in epithelial cell cytoplasm .Experiment show that theexpression of VEGF in the group of ectopic endometrium in ovarian chocolatecysts are higher in the cases with EMs. It is significantly greater than that in thecontrol group (P<0.05).2. eNOS is expressed mainly in epithelial cell cytoplasm .Experiments indicate thateNOS is highly expressed in the group of ectopic endometrium in ovarian chocolate cysts in cases with EMs. In the control group, endometrial eNOS isalso has a certain expression, but significantly lower than the experimental group.There were highly differences between the two group (P<0.05).3. In the expression of VEGF and eNOS in endometriosis coming from 30 patientswith EMs, both VEGF and eNOS have a strong expression. The analysis of rankcorrelation showed that both of them were correlated (r=0.747, P<0.001).ConclusionBoth VEGF and eNOS have a higher expression in ectopic endometrium inovarian chocolate cysts. They have the relevance, implying that both VEGF andeNOS have the relationship with the development of EMs and may play an importantrole.
