Object i V6: Using Dii marking the hepatocytes before intrasplenic hepatocyte transplantation ( HT ) to observe their migration and surviving morphology, then to confirmed these with EMA immunohistochemistry staining;and also to detect their physiologic function and proliferation with PAS staining,PCNA immunohistochemistry staining. Methods Rat hepatocytes were harvested according to the liver purfusion method of Seglen. After staining with Dii, the hepatocytes were injected into the parenchyma of recipient~s spleen. Rats were killed at 3,7,14,30,60,100 days after HT. The cryosection and paraffin section samples of liver, spleen,lung,heart,kidney,and brain were observed under fluorescence microscopy, also under light microscopy after PAS staining ; EMA,PCNA immunohistochemistry staining. Resu Its : Hepatocytes were well marked by Dii. Through tracing study,we found hepatocytes survived not only in liver,spleen and lung but also migrated into main organs,such as heart,kidney,and brain. These results were confirmed with EMA immunohistochemistry staining. These cells appeared normal morphology, expressed hepatic glycogen synthesis and nuclear proliferation. Conc I us i ons Dii is new ,facilitated and reliable marker used in tracing study. Intrasplenic transplanted hepatocytes can immigrate and survive in the heart ,kidney and brain. They can also express function and proliferation.
