[Objective] To evaluate the effect of indoleamine 2,3-dioxygenase on fuction of transplanted hepatocytes in vitro and in vivo test.[Methods] BABL/c mice hepatocytes were transfected with recombinant adeno-virus IDO gene. Mice hepatocytes were isolated by the modified two-step method described by Seglen. The viability were assessed by trypan blue exclusion. In vitro,cultured the recombinant viruses with hepatocytes, monitored the mRNA and protein expression levels by RT-PCR and Western Blot,respectively. BABL/c mice were divided into 5 groups, which is the hepatocyte group, ido group, CsA group,IDO+1-MT group and Shell adenovirus group,respectively. Hepatocytes from the five groups BABL/c mice were cultivated commonly with C57BL/6 spleen T lymphocytes in vitro, respectively. The concentrations of ALT and LDH in the supernatant in different cultural period (1,2,3,4,5days) were examined with ELISA kit and compared respectively. Hepatocytes from the five groups BABL/c mice were transplanted into C57BL/6 spleen in vivo. The concentrations of ALT and LDH in the spleen homogenate in different cultural period (1,3,5,7,10days)were examined with ELISA kit and compared respectively.[Results] Hepatocytes effectively expressed IDO after transfection. Experiments In vitro and in vivo have shown that the concentrations of ALT and LDH in IDO group was obviously lower than the hepatocyte group and IDO+1-MT group in different period (days after surgery) (P<0.05), While the concentrations of ALT and LDH in IDO+1-MT group was no lower than the hepatocyte group(P>0.05).[Conclusion] IDO to transplanted hepatocytes has protective effect.
