Purpose:To investigate the significance of fried turmeric affect to expression of nuclear transcription factor-κB (NF-κB) and dihydronicotinamide adenine dinuclectide phosphate (NADPH) of the rats after vascular injury for the prevention of restenosis after PCI to provide experimental basis.Material and method:Analog PCI operation to cause acute vascular intimal injury, in which turmeric group 36 and saline group 36 and 36 with the sham group and 36 normal group as control. Turmeric sets of the general routine PCI after turmeric (100mg/Kg/d) administered orally; saline treated group after the general conventional analog PCI saline 3 ml / d orally; sham laparotomy suture does not insert the balloon catheter; normal control group did not use any intervention. PCI simulation at 3,7,14 days after the first use of light and electron microscopy changes in the structure of rat endothelial cells;detected by ELISA in 144 rats serum the NF-κB, NADPH expression.Results:1. Light microscope: see turmeric fried group 7 days after injury medication 7 days compared with the model group of local endothelial vascular injury and intimal damage off a slight narrowing, the degree of local vascular stenosis visible reduced. And normal control group, but there are still differences, the lumen area was significantly smaller than the control side without injury.2. Electron microscope: the third day after balloon injury seen in the endothelial cells of irregular columnar, cell shrinkage, cytoplasmic stain, nuclear fragmentation. See the release of a small number of apoptotic bodies. Seventh day, cell necrosis increased. Cells in model group was significantly necrosis, there are a large number of lipid and necrosis of fat soluble substances, and necrosis of fried turmeric and other cells was significantly reduced compared with the control group, only see a small amount of cells debris. 3. In the first 3 days of drug intervention: turmeric fried and model group (NS) of serum levels decline in NF-κB (P <0. 05); turmeric fried and model group (NS) a decrease of serum levels NADPH trend (P> 0. 05), compared with the sham group no significant differences (P> 0. 05).4. In the first 7 days after drug intervention: turmeric fried and model group (NS) NADPH content of serum and NF-κB was significantly decreased (P <0. 05); compared with the sham group decreased serum NADPH, (P > 0.05).Conclusion:1. By light and electron microscopy of the simulated PCI vascular endothelial cells after changes in the structure of the observations, indicating that turmeric fried protective effect on vascular endothelial clearer.2. NF-κB and NADPH may be involved in restenosis after PCI pathophysiological changes. Early treatment may inhibit NADPH activity of turmeric, significantly reduced the level of NF-κB, reduced NF-κB/IκB complex phosphorylation, inhibition of vascular smooth muscle proliferation and neointimal formation and protection of endothelium.3. Detection of serum levels of NF-κB and NADPH levels after vascular injury may be helpful in assessing the degree of stenosis and outcomes.4. Turmeric fried rat NF-κB and the regulation of NADPH level has a certain role.
