Rheumatoid arthritis (RA) is a chronic, systemic, autoimmune disease characterised by symmetrical polyarthritis in clinic. The main pathological change is persistent proliferative synovitis that can lead to irreversible damage of joint cartilage. These factors contribute to functional limitation, disability, comorbidities, and reduced quality of life. All ages can develop RA, now it is thought a disease which is hard to treat. Till now, there is not ideal therapy for RA except hormon and immunosuppressive agents. While Chinese medicine has been proved in clinic that it is effective and has few side effects, so it becomes a research focus. In this study, we duplicated mice model of RA, and investigated effects and mechanism of SangGuiTongBiJi on treating RA through histological observation by using immunohistochemical and immunofluorescence method.ObjectiveStudy the effects of SangGuiTongBiJi on treating mice with RA, to make the foundation for further research.Methods1. Duplicate mice model with RA Model mice were randomly divided into four groups: model group, methotrexate treatment group (MTX group), Glucosida Tripterygii TOTA treatment group (T2 group) and Chinese medicine treatment group (SangGuiTongBiJi group). Normal mice were uesed as control. After four weeks treatment, all mice were executed. Arthritis Index (AI) was assessed and morphological changes of joints were observed.2. Observe expression of MMP3 and VEGF in Synovial tissue of joint by immunohistochemical method.3. Observe expression of MIP by immuno fluorescence method.Results1. Model mice developed significant joint swelling, the number of involved joint in all treatment groups was less than model group. AI in treatment groups also smaller than model group.2. Control group showed normal ankle joint structure. Model group demonstrated corpora mammillaria synovium proliferation accompanied by lymphocytes infiltration in articular cavity and bursa mucosa. Necrosis of cartilage, blood vessel dilation, pannus also be observed. All changes mentioned above markedly alleviated in MTX group, T2 group and Chinese medicine group.3. There was low expression of MMP3 and VEGF in normal mice, and no MIP expression was found in normal group, but the expression of MMP3, VEGF and MIP significantly increased in model group. After treatment, expression of these three kinds of protein decreased.Conclusion SangGuiTongBiJi can reduce the number of involved joints and severity of pathological changes in model group, its treatment mechanism may through depressing the expression of MMP3, VEGF and MIP.
