Tourette syndrome is an inherited neuropsychiatric disorder with onset in childhood, characterized by multiple physical (motor) tics and at least one vocal (phonic) tic; these tics characteristically wax and wane. Tourette's is defined as part of a spectrum of tic disorders, which includes transient and chronic tics. The severity of the tics decreases for most children as they pass through adolescence, and extreme Tourette's in adulthood is a rarity. The exact cause of Tourette's is unknown, but in recent years the immune aectiology of TS is gradually concerned by researchers. Western medicine concentrate more on dopamine receptor blocker, by using monoaminergic antagonist, central alpha receptor stimulating agent and antiepileptic drug treatment. The short-term clinical effect is acceptable, but with high recurernce rate and great side effect. Compared to western medicine the TCM therapy has better clinical effect in curing TS and easily accepted by the parents. Professor LiuBiChen from pulmonary theory invented Xifeng Jingning Tang for clinical treatment, it is one of the effective prescriptions for TS. For better explore of honored TCM doctor experience in treating complicated-syndrom, we start from the immune perspective to further understand the use of Xifeng Jingning Tang in the TS' treatment mechanism. The topics include two parts of literature research and experimental research.1. Literature research is discussed from three aspects:①Mainly discuss TCM literature related-knowledge of TS;②Primarily introducing the western medicine knowledge of TS;③mainly discuss the existing literature on Xifengjingning tang.2. Experimental research:Objective:To observe Xifeng Jingning Tang's effect on immune function of Immunodef icient mice and to discuss Xifeng Jingning Tang mechanism in treating TS.Methods:The experiment began with blank controller. The model group are equally injected with 20ml·kg-1 of 0.9% sodium chloride, Thymosin group injected with 15mg·kg-1 Thymosin, Xifeng Jingning Tang small, medium and high dose are set respectively by 3g·kg-1,6g·kg-1,12g·kg-1. Above groups were intraperitoneal injected on daily basis 20ml·kg-1 for 7 consecutive days. On day 2, depilatory agent was coated on mice abdomen. On day 3, the blank controller group was intraperitoneal injected with 0.9% sodium chloride 0.1 ml·10g-1 daily, the others are injected with cyclophosphamide 50mg·kg-1 for 3 consecutive days. At the same day, using 1% dinitrofluorobenzene (DNFB) 25ul coated onto mice abdomen, intensively on the next day. After 4 days of sensibilization, the mice's right ear was coated with DNFB 10ul. After 24h, the mice were killed and both ears were taken using 9mm punch swollen ear tool for weighed using electronic scale.Results:①Mice's ear swollen degree on model group was significantly lower than blank controller group (P<0.01);②Thymosin group and high dose group of Xifeng Jingning Tang swollen degree was significantly higher than the model group (P<0.01);③Xifeng Jingning Tang medium dose group of swollen degree was also higher than the model group (P<0.05);④Xifeng Jingning Tang small dose group have no significant difference compared to the model gorup.Conclusion:①Creation of the experiment model were successful;②Xifeng Jingning Tang is effective against immune suppression to reduce the degree of swelling in mice's ear, can also enhance immune response to suppress the level of DTH in mice;③Immune regulation may be one of Xifeng Jingning Tang mechanisms in treating TS.
