Experimental Studies Of Ethanol-type Gastric Ulcer Rat Model Of Sleep Time And Acupuncture Effects

The gastrointestinal disease affects sleep. This viewpoint can find on the book "the yellow emperor of internal medicine", it is said "the stomach is lying with unease". In TCM theory, the stomach is well when it is descending. If it fails to descend, then block the defending Qi result in insomnia. The gastric ulcer is one of the global diseases. The clinic manifestations are rhythmicity, repeatedly and longtime duration of upper abdominal pain, accompany the stomach and intestinal symptoms of nausea, vomiting, belching, acid regurgitation. In TCM theory, it belongs to "stomachache", "acid regurgitation" and "fullness". Using the western therapy still remain problems which are hardly to cure and easily to occur. After one year stop taking the regular acid resistant treatment for the ulcer, there is 60% recurrence rate. The acupuncture treatment can protect the gastric mucosa through many approaches. The most advantage is avoiding the side effects by the oral medicine and the decreased absorbing of medicine due to the digestive disease. Therefore, searching a proper, effective acupuncture method is one of the key projects.ObjectiveFrom experiment research, choose the alcoholic ulcer rats as the objects, observe the rat's gastric mucosa pathological and morphological changing, take the dopamine which is the important gut peptides as the major target, to study probable mechanism of the acupuncture cure the gastric ulcer, and then explore the correlation between the stomach and sleep, in order to apply some evidence for the clinic.Method①Using the absolute alcohol to irrigate stomach, the rats are random divided into five groups.②Observe the rats'free activities and record the sleep time.③According to the Guth scoring method to measure the ulcer, calculate the injury index number, and using the light microscope to observe the stomach pathomorphologyResult1. The number of the free walking:the model group compare with the other group, P<0.01, very significantly different; the blank group compare with the treatment groupⅠandⅡ, P< 0.05 significantly different; the treatment groupⅠwithⅡ, P< 0.05 significantly different2. Sleep time:the model group compare with treatment groupⅠandⅡ, P <0.05 significantly different; the model group compare with treatment groupⅢ and blank group, P< 0.01 very significantly different. Between all the treatment group, without significantly different.3. Ulcer damage index score:the model group compare with treatment groupⅠandⅢ; P< 0.01 very significantly different; the model group compare with treatment group; P<0.05 significantly different; the treatment groupⅢwithⅡ, P<0.01 very significantly different.4. Dopamin:the DA in blood serum shows that model group compare with the others, P<0.01 very significantly different; the treatment group III with blank group and model group, P<0.05 significantly different. The DA in corpus striatum shows model group compare with blank group, treatment groupⅠ, P＜0.05 significantly different; compare with treatment groupⅠ, P<0.05 significantly different.Conclusion1. The acupuncture can shorten the ulcer area and decrease the ulcer index, promote the stomach mucous recover which has evidence effect for curing gastric ulcer.2. The rats with gastric ulcer, their pentobarbital sodium induced sleeping time are shorter.3. The acupuncture with pentobarbital sodium have prolong the rat's sleeping time, which indicates the acupuncture therapy can improve the gastric ulcer rats' sleep.4. The acupuncture can regulate gastric ulcer rats' DA contents in the serum and corpus striatum5. The mechanisms of acupuncture enhance the healing rate and improve sleep may through regulate DA contents in the serum and corpus striatum.
