Alcoholic liver disease is caused by long-term alcohol abuse. The objects of this study include two aspects:on the one hand,t he target of our study is the compatibility of Flos puerariae and seeds of Hovenia dulcis Thunb,as the same time we need to discuss the compatibility of traditional Chinese medicine theory that modern science support from the pharmacodynamic point of view; on the other hand, the contents of the study is the alcoholic liver disease and the study can expand clinical application of the compatibility of Flos puerariae and seeds of Hovenia dulcis Thunb and enrich the research of the compatibility of Flos puerariae and seeds of Hovenia dulcis Thunb to prevent and treat the acute and chronic alcoholic liver disease disease.The alcoholism rats model were induced by administrating wine. Drug is extracted by ethanol(mainly the group of Flos puerariae and seeds and the Flos puerariae in the group of the compatibility of Flos puerariae and seeds of Hovenia dulcis Thunb)and water. We need to observe some aspects that reflect the effects of the drug on the alcoholic liver disease. We use tissue morphology, biochemistry, enzyme histological techniques to detect the effect of the drug and compare which drug is better to the alcoholic liver disease.The indicators and results of this study include the following aspects:①The effect of the compatibility of Flos puerariae and seeds of Hovenia dulcis Thunb on liver index of rats with alcoholic liver diseaseIn one experiment,the drug is extracted by ethanol. The liver index of the model group increased significantly (p<0.001) compared with the control group. The liver index of the group of 2:1 is significantly higher than the model group. The other treatment groups have no significant difference compared with the model group.In the other experiment, the drug is extracted by water. In this experiment, The liver index of the model group increase significantly compared with the control group and the index of the group of Flos puerariae, seeds of Hovenia dulcis Thunb and 1:2 are significantly lower than the model group,as the same time, the liver index of the group of 2:1 is significantly higher than the group of Flos puerariae,and seeds of Hovenia dulcis Thunb②The effect of the compatibility of Flos puerariae and seeds of Hovenia dulcis Thunb on ALT,AST,ALPIn one experiment,the drug is extracted by ethanol. In this experiment ALT of the model group increase significantly compared with the control group, but all the treatment group have no significant difference compared with the model group. AST of the model group has no significant difference compared with the model group and all the treatment group have no significant difference compared with the model group. ALP of the model group is significantly higher than the control group and the ALP of all the treatment group are significantly lower than the model group.In the other experiment, the drug is extracted by water. ALT of the model group increases significantly compared with the control group. ALT of the Flos puerariae and seeds of Hovenia dulcis Thunb group is significantly lower than the model group, ALT of the other treatment goups have no significant difference compared with the model group. AST of the model group increases significantly compared with the control group. In all treatment groups, except for positive drug Dongbao Gantai group, other groups show statistical differences compared with the model group but we can not find which group shows relatively good results between compatibility groups and the single herb treatment group. ALP of the model group has no significant difference compared with the control group. In all the treatment groups, except for positive drug Dongbao Gantai group, other groups have decrease trend. As the same time, ALP of 1:2 group is significantly lower than the model group and has a significant difference.③The effect of the compatibility of Flos puerariae and seeds of Hovenia dulcis Thunb on TP,Alb,TGIn one experiment,the drug is extracted by ethanol. TP of the model group has decrease trend but has no significant difference compared with the control group. TP of all the treatment groups has no significant difference compared with the model group. Alb of the model group has no significant difference compared with the control group. In all the treatment groups, Alb of the 1:2 group is higher than the model group and has a significant difference. As the same time, Alb of the 1:2 group is significantly higher than the group of the Flos puerariae. Alb of the seeds of Hovenia dulcis Thunb group is significantly higher than 1:1 group. Alb of other group has no significant difference compared with the model group. TG of model group is significantly higher than the control group. TG of the positive drug Dongbao Gantai group and the Flos puerariae group are significantly lower than the model group.TG of other groups has no significant difference compared with the model group.TG of the 1:2 and 2:1 group are significantly higher than the Flos puerariae group and the seeds of Hovenia dulcis Thunb group.In the other experiment, the drug is extracted by water. TP of model group is significantly lower than the control group. In all the treatment group, except for positive drug Dongbao Gantai group, TP of other groups shows significantly higher than the model group and has statistical difference. TP of all the different ratio compatibility groups is significantly higher than the Flos puerariae group and the seeds of Hovenia dulcis Thunb group. Alb of the model group is significantly lower than the control group. Alb of All the treatment groups has no significant difference compared with the control group. TG of the model group is significantly higher than the control group. TG of all the treatment groups is significantly higher than the model group and has statistical difference.④The effect of the compatibility of Flos puerariae and seeds of Hovenia dulcis Thunb on SOD,MDA,GSHIn one experiment,the drug is extracted by ethanol. SOD of the model group is significantly lower than the control group. In all the treatment groups, SOD of Dongbao Gantai group and the Flos puerariae group is significantly higher than the control group. SOD of the other groups has no significant difference compared with the model group. SOD of 1:1 and 1:2 group is significantly lower than the Flos puerariae group. MDA of the model group is significantly higher than the control group. In all the treatment groups, MDA of the seeds of Hovenia dulcis Thunb group,1:2 and 2:1 group is significantly lower than the model group and MDA of the 1:2 and 2:1 group is significantly lower than the Flos puerariae group. MDA of other groups has no significant difference compared with the model group. GSH of the model group is significantly lower than the control group. GSH of all the treatment groups has increased trend but has no significant difference compared with the model groupThe experiment showed that in the water-extracting Chinese medicine part, SOD activity in rat liver of the model group increased significantly (p<0.001) compared with the control group. SOD activity in rat liver of the group of Flos puerariae and the group of 2:1 is significantly higher than the model group, the others groups trended to increase compared with model group, but no significant differences. The SOD activity in rat liver of both Flos puerariae and seeds of Hovenia dulcis Thunb 1:1 group and 2:1group is significantly higher than the Single seeds of Hovenia dulcis Thunb group. The content of MDA in rat liver of the model group is significantly higher than the control group. The others groups is lower than the model group, The content of MDA in rat liver of Flos puerariae and seeds of Hovenia dulcis Thunb 1:2 group and 2:1 group are is significantly higher than the Single seeds of Hovenia dulcis Thunb group, and there is significant statistical differences. The content of GSH in rat liver of the model group is significantly lower compared with the control group. the others groups trended to increase compared with model group, but no significant differences ⑤The effect of the compatibility of Flos puerariae and seeds of Hovenia dulcis Thunb on the alcohol density in serum and the activity of ethanol dehydrogenase(ADH)in liver with alcoholic liver diseaseIn the experiment of ethanol-extraction Chinese medicine part, the concentration of alcohol in rat blood of all the treatment groups is significantly lower than the model group except the Flos puerariae group, and the concentration of alcohol in rat blood of Flos puerariae and seeds of Hovenia dulcis Thunb 1:1 group and 2:1 group is significantly lower than the Single seeds of Flos puerariae group, and there is significant statistical differences. The other treatment groups trended to reduce but no significant differences compared with model group.The activity of ethanol dehydrogenase(ADH)in liver of contral group and other treatment groups has no significant statistical differences compared with model group.In the experiment of water-extracting Chinese medicine part, the concentration of alcohol in rat blood of all the treatment groups is significantly lower than the model group, and there is significant statistical differences. In addition, The concentration of alcohol in rat blood of both Flos puerariae and seeds of Hovenia dulcis Thunb 2:1 group and 1:2group is significantly lower than the Single Flos puerariae group and the Single seeds of Hovenia dulcis Thunb group. The activity of ADH in liver of model group is significantly lower than the contral group, and The activity of ADH of all the treatment groups is significantly higher than the model group. The activity of ADH in liver of Flos puerariae and seeds of Hovenia dulcis Thunb 2:lgroup is significantly higher than the Single seeds of Hovenia dulcis Thunb group. And The activity of ADH in liver of Flos puerariae and seeds of Hovenia dulcis Thunb 1:2 group is significantly higher than the Single seeds of Hovenia dulcis Thunb group and the Single Flos puerariae group.⑥The study on the liver pathologic changes of different proportion of Flos puerariae and seeds of Hovenia dulcis Thunb with alcoholic liver diseaseLiver pathologic changes with alcoholic liver disease of the rat are basically the same in water-extracting part and ethanol-extracting part. As a whole, the healing effect of Flos puerariae and seeds of Hovenia dulcis Thunb 2:1 group is the best in all the treatment groups⑦The study on the hepatic Ultrastructure of different proportion of Flos puerariae and seeds of Hovenia dulcis Thunb with alcoholic liver diseaseIn the experiment of ethanol-extraction Chinese medicine part, cell nucleus,mitochondria,endoplasmic reticulum in the liver of the rat were seriously damaged of the model group and has a large number of fat cells but all the treatment groups is alleviated accordingly. In all the treatment groups, the group of 2:1 has the strongest protective effect to the liver cell ultrastructure.We can summarize the following points:①from the overall biochemical indicators, we know that different ratio compatibility groups have better effects than the Flos puerariae group, seeds of Hovenia dulcis Thunb group on the protection of liver cells that in the rats with alcoholic liver disease;②from all the results,we know that drug extracted though water have better effects than alcohol extraction;③from the overall biochemical indicators,whether the drug is extracted though water or alcohol,we can not summarize which group is best.We believe that the drug can improve the the activity of alcohol dehydrogenase.So ethanol is metabolize into H2O and CO2 though the pathwaw of alcohol dehydrogenase.This way can reduce the production of acetaldehyde and free radical that produced though the pathway of microsomal ethanol oxidizing system(MEOS) and catalase system.
