Pu'er Tea Joint Caloric Restriction Antitumor Experimental Research

Objective:To study the anti-cancer ability of Pu-er tea, Green tea and Black tea,using breast cancer rat model,combined with energy restriction treatment,and investigate possible pathway involved.Methods:1,The inhibition effect of calorie restriction on breast cancer and colorectal cancer is the best among all kinds of cancer. At the same time,Pu-erh tea can remarkably induce breast cancer cells growth arrest:treating murine colorectal cancer cells with 1640 culture medium containing different concentrations of Pu-erh tea, and perform a MTT test,to see whether there is inhibition of Pu-erh tea on colorectal cancer, if so, the colorectal cancer rat model can be set up to be carried out in animal experiments.2,Pu-erh tea combined caloric restriction treatment on experimental animal in curing studies:breast cancer model rats, feeding Pu-erh tea or water, at the same time they were divided into calorie restriction group and the control group, their body weight were weekly weighed, changes in tumor volume of rats were measured, recorded the survival rate of the rats, at the end of treatment rats'liver weight, kidney weight, spleen weight, etc. were weighed, biochemical detection of liver and tumor tissue,such as HE biopsy, RT-PCR and immunohistochemical analysis,were performed against the key enzymes in glucose and lipid metabolism, comprehensive study were done to evaluate the anticancer activity of these two treatments and explore the possible tumor suppression pathway.3,Pu-erh tea combined caloric restriction treatment on experimental animal in prevention studies:the rat model of breast cancer was also used, just before the rat model was set up, a month of Pu-erh tea and energy restriction treatment was given to normal rats, after that continue to give these two treatments for another month, same tests of step 2 were performed, so were comprehensive study and evaluation on their tumor suppression role in cancer prevention and exploring its possible tumor suppression pathway.4,Comparison of the antioxidant activity of Green tea, Black tea and Pu-erh tea: ORAC analysis5,Green tea, Black tea and Pu-erh tea combined caloric restriction comparative study on tumor suppression activities (ibid.2 and 3, including treatment and prevention experiments):rat model of breast cancer, before and after the beginning, were given the same concentration of green tea, black tea, Pu-erh treatment, combined with energy restriction, body weight were weekly weighed, measuring changes in tumor volume in rats, recorded the survival of the rats, weighed liver weight, kidney weight, spleen weight, etc.after the end of treatment, the key enzymes of glucose and lipid metabolism were detected using RT-PCR and immunohistochemical analysis, comprehensive study was performed to evaluate tumor suppression effects of these three kinds of tea and explore its possible pathway.Results:1,The murine colon cancer CT-26 cells growth did not get inhibited by treating with Pu-erh tea.2,Fed with 2.0g/kg body weight per day of Pu-erh tea and combined 30% calorie intake limitation, at a certain extent, tumors were prevented, inhibited, and reduced and survival rat of tumor rats was increased. The effect of prevention studies is better than that of the treatment study.3,Fed with 2.0g/kg body weight per day of Pu-erh tea and combined 30% calorie intake limitation treatment can induce a significant weight reduction in rats, lower blood fat, blood sugar and so on, biopsy results showed that the tumor cells turned to apoptosis, maybe a key enzyme in glucose and lipid metabolism was affected:acetyl coenzyme A carboxylase (ACC), or fatty acid synthase (FAS).4 Comparison of anti-oxidantion activity of three kinds of tea:green tea> pu-er tea >black tea.Comparison of prevention and suppression of tumor growth:Pu-erh tea>Green tea>Black tea.Conclusion: 1,Pu-erh tea combined calorie restriction can play better the effect of preventing cancer, inhibiting tumor growth and prolong survival in rats, significantly improving the living condition of the tumor in rats.2,Overall,the prevention group is better than treatment group.Long-term consumption of tea can be seen, but the degree of eating on the prevention of cancer and prolonging life have very good results.3,Tea and sugar metabolism of tumor suppressor mechanism may be related to its pathway key enzyme at the protein level and RNA level are significantly inhibited, the United calorie restriction, suppression better.4,Green tea, black tea, Pu'er tea has some of the pre-prevention of cancer and inhibit tumor effect, although the antioxidant activity of green tea the best, but the synthesis is concerned, the tumor suppressor effect of tea is the best, green tea, followed by good tea may inhibit the glucose metabolism pathway.
