RA has a high incidence ratio in China (0.32-0.36%), which has been one of the main reasons that people lose labour force and become disabled. Its modern medical cause is not clear, and there are no cure method and preventive measures. Recent clinical and experimental research show that Chinese medicine treatment of this disease have made great advances, especially the acupuncture therapy which has certain effect, little side effects, is becoming an important methord, and getting more and more attention. With the in-depth research in RA experiment, some drug (such as tripterygium wilfordii) was found had certain curative effect in the treatment of this disease,but its poison effect influence its long-term applications, so taking drugs alone can not obtain ideal curative effects. Therefore, it is worth attention that if combine the needle and medicine can get complementary advantages, reduce toxicity, increase its effect and achieve the desired therapeutic effect. There is little research of combining electro-acupuncture and tripterygium wilfordii, therefore, it is a valuable project to observe the curative effect and discuss the mechanism.In this paper, we select the model of adjuvant arthritis (AA) rats, focusing on IL-1β; TNF-αand IL-10 as the breakthrough point, to observe the curative effect of combining electro-acupuncture and tripterygium wilfordii. In the experiment,50 male SD rats were randomly divided into 5 groups as follows:blank control group (n=10), model group (n=10), electro-acupuncture in Zusanli(ST36)group(n=10), combine electro-acupuncture in Zusanli(ST36) and tripterygium wilfordii group (n=10) and tripterygium wilfordii group (n=10). Observe the behavioral tests such as:the weight, the AI and volume of the ankle. Inspect the level of IL-1,TNF-a and IL-10 in blood serum. Combined with histological sections by optical microscope and inspect the level of uric acid, urea nitrogen, serum creatinine, cereal third transaminase, aspertate aminotransferase and albumin in blood serum. Compare different intervention methods for chronic AA rat models in the influence of inflammatory cells and anti-inflammatory repair treatment, the different degree of liver and kidney damage, to evaluate and analyse comprehensively. The experimental result shows:1. Each intervention methods have curative effect in control arthritis symptoms, it can be reflected in the acute phase and long-term curative effects observtion. The difference between groups in acute period is not very significant, but electro-acupuncture in Zusanli (ST36) and tripterygium wilfordii group has the best curative effect in the long run.2. Three intervention methods can reduce the level of IL-1 and TNF-α, increase the consistence of IL-10 back to normal levels (P<0.05), there is no statistically significant differences (P>0.05). Three intervention methods have benign induction on the inflammatory cytokine level, stimulative anti-inflammatory repair response and immune response. The electro-acupuncture in Zusanli(ST36) and tripterygium wilfordii group has the best curative effect by comparing.3. Compared with model control group, the phenomenon such as:inflammatory cell infiltration and vascular proliferation was improved. The electro-acupuncture in Zusanli (ST36) group and combining electro-acupuncture in Zusanli(ST36) and electro-acupuncture in Zusanli(ST36) group were better than tripterygium wilfordii group4. Uric acid:only electro-acupuncture in Zusanli(ST36) and tripterygium wilfordii group has no difference with normal group; the level of other two group was higher than model control group. Alanine aminotransferase and aspertate aminotransferase:The electro-acupuncture in Zusanli(ST36)group and electro-acupuncture in Zusanli(ST36) and tripterygium wilfordii group had significant differences with model control group, (P<0.05), and the two had no significant differences with normal control group. Combining electro-acupuncture in Zusanli (ST36) and tripterygium wilfordii group had the best effect in reducing the liver and kidney damage.Conclusion:the methord of combining electro-acupuncture in Zusanli(ST36) and tripterygium wilfordii to treat rheumatoid arthritis, can get complementary strength, collaborative curative effect and reducing toxicity, it is worth extending.
