Objective:To discuss the treatment effect and the impact on liver function andreproductive system of different doses Tripterygium wilfordii compoundrecipe gel (TWCG) for external use on adjuvant arthritis(AA) in rats.Methods:Rats were percutaneous administrated with homemade different dosesof TWCG after the freund’s adjuvant arthritis model was established. Thegambrel swelling degrees were evaluated;the serum levels of tumor necrosisfactor-α(TNF-α) and interleukin-4(IL-4)were detected by ELISA;thelevels of matrix metalloproteinase-3(MMP-3)in ankle joint cartilagewere determined by immunohistochemical method; The serum levels ofalanine aminotransferase(ALT),aspertate aminotrans-ferase(AST)were tested with the automatic biochemistry analyzer to observe thehepatotoxicity; the levels of vascular endothelial growth factor(V EGF)inovary were determined by immunohistochemical method to observe the reproduction toxicity.Results:The gambrel swelling degree in TWCG groups and voltaren group weresignificantly reduced(P<0.05);the levels of TNF-α in TWCG groupsand voltaren group were visibly decreased and IL-4were visiblyincreased(P<0.05);the content of MMP-3in ankle joint wassignificantly reduced. Effects in TWCG high dose group and median dosegroup were better than voltaren group; however,the differences of ALT andAST were not statistically significant among the groups;the expressionlevels of VEGF in rats ovaries had no statistical difference among thegroups.Conclusion:TWCG had obvious anti-inflammatory and detumescence effect on AArat.It could protect cartilage tissue effectively, while it had no significantliver toxicity and reproduction toxicity.
