The sprawl of counterfeit and substandard drugs has seriously affected the healthy development of national medical projects. Since 2003, the National Institute for the Control of Pharmaceutical &Biological products (NICPBP) has arranged a specialty research group, focus on using modern chromatography and spectrum analytical technology to develop and study the rapid drugs testing system. NIR has been widespread used to inspect the counterfeit and substandard drugs for its rapid test,non damage to sample. The key point of establishment drug's NIR modules for qualitative and quantitative was selection of characteristic spectral bands. However, It was very difficult to exactly make structure resolution for NIR spectrum mostly containing compound double frequent or combine frequent absorb peaks produced by-H groups such as O-H,N-H,C-H,S-H end so on. So, this selection is largely based on experience in courrently, and there is not effective method to verify the characteristic representativeness between NIR absorb peaks and main structure unit. Here we reported a new method for selecting characteristic spectral bands:using the generalized 2D correlation spectroscopy as reference, we analyzed the IR-NIR 2D correlation spectra of chemical compounds. By the virtue of unambiguous functional groups of IR spectral and by the correlation analysis of IR-NIR spectrum, we identified the typical characteristic wave number regions that were closely related to the structure of chemical compounds and relatively less disturbed as the characteristic spectral bands in the NIR. This study was not only providing a new mentality and method to resolve the problem of structure resolution about NIR and verification the selection of characteristive spectral bands in NIR modules but also improving the ability of detection in complicated system. In this study, frist, we explored how to establish the 2D-IR-NIR correlation spectroscopy by survey external perturbation and how to pretreat data, conclude genenal process and principle. Second, with illegal additives Sildenafil, Tadalafil, Metformin hydrochloride and comtermporary additives Sildenafil-Tadalafil compound sample as an example, we explored how to utilize the 2D correlation spectroscopy to select the characteristic spectral bands, thereby establishing a rapid method for screening illegal additives in Chinese patent drugs and healthcare products. Finally, by comparing with 6 inject for cephalosporin,l inject forβ-lactam simple preparation and 1β-lactam compound preparation, we examined the feasibility of our method for selection and verification modeling spectral bands in NIR modules.
