Cold And Heat Effects Of The Medicine The Objective Differences Preliminary Study Shows The Difference Method Of Chinese Medicine Based On The Hot And Cold Plates

The description and differentiation to Cold and Hot natures, the main Drug Nature of Chinese medicine, has been all along the difficult and hot issue in theoretical researches.In this study, the cold/hot differentiating assay was firstly established in an ethological view of the temperature tropism of animal.The intervention of Red gingseng, American ginseng, Coptis,Mahuangtang and Maxinshigantang were investigated via Methodology. And this paper initially indicated the objective differences of Cold/Hot nature of Chinese medicines,the main results were listed as follows:1.The results of methodology investigation of the cold/hot differentiating assayThe factors such as species,sexes,taming time ect of mice were inspected and the results showed that the experiment animal was KM mice for its sensitivity to temperature, the taming time depend on the settling time of mice to the cold/hot pates(2h～6h), the temperature zone could be setted at the arrange of 15℃～40℃and the ambient tempraure was 20±2℃with relative humidity 60～80%。2.The establishment of constitutional pathologic models (Weak Model and Strong Model)The group of Weak Model was established by restrict food plus swimming, and the group of Strong Model was established by feeding high protein food without limit. The mice of Weak Model group showed the feature of "Deficient Cold symptoms",such as fatigue,poor appearance,stagnant growth in weight, lower viscera index and body temperature, decrease in water, cold limbs and tail etc..The macroscopic behavioral index of remaining rate (RR) on warm pad (40℃)of mice increased significantly (p<0.05),suggesting the enhancement of HOT tropism, meanwhile, the internal indexes of activities of ATP,SDH,T-AOC,SOD,oxygen consuming volume and the specific value of cAMP/cGMP decreased significantly (P<0.05),activity of MDA and the quantity of cGMP increased obviously (P<0.05).suggesting the decapability of energy metabolism and antioxidant ability. While the Strong Model group changed on the inverse way.3.Effect of drugs to cold/hot animal's thermo-activity and energy metabolism Study on animal models showed that the animal's thermo-activity system could separate the difference between Red gingseng and American gingseng, Gingseng and Gingseng fower, Mahuangtang and Maxinshigantang,Coptis and its processed materials.The rate of Red gingseng, Mahuangtang and Gingseng on warm plate increased which suggesting the hot nature of them; While American gingseng, Maxinshigantang and Ginseng flower showed cold nature.In our study, we found that Coptis Chinensis Franch and its prosses materias also showed the cold nature, compared with crude C. Chinensis Franch, the Cold nature of bile-processed C. Chinensis Franch enhanced while the ginger-processed C. Chinensis Franch changed on the inverse way. The changing sequence was consistent to the theoretical prognostication or definition.The oxygen consumption, ATPase activities, total anti-oxidant capability (T-AOC), superoxide dismutase (SOD) activity etc.of mice were examined, It was found that cold drug such as Coptis,American gingseng and Maxinshigantang could inhibit both normal and model animal's energy metabolism; while hot drug such as Red Ginseng, Maxinshigantang could enhance the energy metabolism, suggesting that the changes of temperature tropism and energy metabolism might relate with Zheng hou of animals and cold/hot nature of Chinese medicine,which might be internally regulated by body's energy metabolism induced by the alteration of ATPase activity. In a summay, the the cold/hot differentiating assay might be the better method to distinguish the cold and hot nature of Chinese medicine;The varity temperature tropism of mice after treated with different drug was consisted with the ancient TCM theory;Body's energy metabolism induced by the alteration of ATPase activity might be one of the internal mechanisms;it could be concluded that the difference of human body to adapt cold and hot environment might be a result of intervention of medicines to energy transaction, also might be considered as a kind of expression manner of the Cold and Hot natures of Chinese medicines.
