From the angle of practical application,an instrument system of gene amplification which takes semiconductor heating up and cooling module as the inner core was designed and made up to against the problems of traditional system of gene amplification that large volume,narrow range of temperature-adjusted,bad temperature-controlled,slow velocity of temperature heating up and cooling and unfriendly interface of human-computer.First of all,the technique principle and realized conditions of the polymerase chain reaction(PCR) have been analyzed;according to the specific demands of instrument system of gene amplification,the overall system design program has been proposed.The analysis instrument system of gene amplification took single chip computer M30627FJPGP as the core was developed,and the design method of the systematic software and hardware was given out.Regarding M30627FJPGP as the core of system-control,the controlled quantity has been obtained according to the present module temperature gathered and executing intelligence algorithms;the switch of MOSFET was controlled through PWM to achieve the purpose of controlling the voltage of both sides of the semiconductor wafers to control the module temperature. System equipped with printer and USB interlace and adopted big liquid crystal screen using all Chinese prompt as the human-computer interface offered good operating and displaying interface to users.Secondly,because the controlling-target of instrument system of gene amplification had characteristics of complicated mechanism,obvious nonlinearity, pure lagging behind,model uncertainty,etc.,and the internal traditional temperature-controlled systems of gene amplification analysis instrument mostly set up on certain model foundation that difficultly to guarantee the temperature-controlled precision,the fuzzy controls algorithms was introduced into the traditional temperature-controlled systems of gene amplification analysis instrument to constitute the intelligent control system which using the controller of fuzzy parameter self-tuning PID to adjust the control parameter online and improving the result of controlling. Finally,the system assembling and debugging process and the instrument operating data analyzed according to combine with the instrument characteristics were provided,the factors that influencing the effect of the heating up and cooling of the instrument of PCR and the specific requirements of the instrument of PCR to electromagnetic compatibility were analyzed.Experiments showed that the temperature-controlled precision of the system could be achieved to±0.2 degrees Centigrade,the velocity of temperature heating up and cooling was fast,the performance was stable.The design method of the systematic software and hardware also has certain interrelated significances to other similar high-accuracy temperature-controlled occasions.
