Objective: The anti-thrombosis effect of different extracts of Ocimum Basilicum L.(OBL) was examined in three different experimental thrombosis models, and the effect of the extracts within active ingredients of different polarity was compared to investigate the type of the probable effective constituents and select the best extracting method, then investigated the action of mechanism from both anticoagulation and fibrinolysis aspects. Methods: Three experimetal models in vivo, pulmonary thromboembolism in mice, FeCl3-induced arterial thrombosis and ligation-induced inferior caval thrombosis in rats, were used to examine the anti-thrombotic activity of aqueous and alcohol extracts of OBL by different extracting methods, and the activity of AT-Ⅲ, PC, PLG, t-PA and PAI-1 in plasma were determined by ELISA to evaluate the effect of the drugs on the anticoagulant and fibrinolysis system. Results: Extracts of OBL could increase the survival rate of the mice suffered from pulmonary thrombosis induced by the mixture of collagen and epinephrine significantly, and decreased the weight of the thrombus induced by ligation of inferior vena cava significantly. The ethanol extract of OBL by supersonic extraction is the most effective of all. The concentration of AT-Ⅲwas decreased, the concentration of PC was increased; the activity of PLG and t-PA were increased, the activity of PAI-1 was decreased slightly, the ratio of t-PA/PAI-1 was increased. Conclusion: The extracts of OBL have strong anti-thrombosis effect, and the fundamental effective components are probablly some lipid soluble flavonoids of relative low polarity, and the action of mechanism may have correlation with increasing of the activity of the anticoagulant and fibrinolysis system.
