Background and ObjectiveEpilepsy(EP) is a kind of chronic brain disease,caused by abnormal synchronous neuronal discharge.It is characterized by transient dysfunction of central neuron system.Although most epilepsy can be controlled effectively by appropriate antiepileptic drugs(AEDs),about 20-25%of them show resistance to AEDs,which is called refractory epilepsy.Temporal lobe epilepsy(TLE) is the most common epilepsy in clinics.Studies on mechanisms of epilepsy before most focus on axon sprouting and synaptic reconstruction in hippocampus.The mainly pathologic characteristics of epilepsy is axon sprouting and synaptic reconstruction which play an important role on TLE.But how they reach to target area accurately is another hot spot.Axon guidance factor is a protein which plays a guiding role on axon growing.There are four kinds of them have been found at present,and they are ephrins,slits,netrins and semaphorins.They control the growth of axon through four mechanisms and guide the axon growing to a certain direction.At seizures,the axon guidance reduced,and the expression of GFAP and GAP-43 increased at the same time which is good to axon sprouting.However,most of the current studies are in vitro or animal model experiments,and few are to epilepsy patients.To reveal the role of axon guidance factor in epilepsy,the expression of EphrinA2 and Semphorin3A in the experiment Materials and Methods12 temporal lobe specimens were from patients with refractory epilepsy in experimental group and 2 temporal lobe specimens are from normal people in control group.We took immunohistochemical test to observe the different expression of EphrinA2 and Semphorin3A between the brain tissue of patients and normal,we used HPIAS-1000multimedia color pathological Graphic Determination of the report management system to deal with the results,and used optical intensity value to marker positive density.All the data were analysis by SPSS10.0 statistical package and the difference was compared with columns.Results1 In experimental group,EphrinA2 was positive in temporal lobe.The volume was larger than that of control group and the formation was round or oval.It was full of Brown-yellow particles.The volume of control group was not expanded and cytoplasm is blue.2 In experimental group,Semphorin3A was positive in temporal lob.The volume of cells was larger than that of control group and the formation was round or oval.It was full of Brown-yellow particles.The volume of control group was normal and cytoplasm is blue.3 The expressions of EphrinA2 and Semphorin3A were higher in experimental groups and these were lower in control groups.4 The optical density value in experimental group had significant difference compared with the control group.(P<0.05)Conclusions1 EphrinA2 plays a promoted effect for neuron sprouting and synaptic reconstruction in patients with temporal lobe epilepsy.2 Semphorin3A plays an inhibitory effect on axonal sprouting and synaptic reconstruction in neurons of patients with temporal lobe epilepsy.3 Maybe EphrinA2 and Semphorin3A are antagonistic in synaptic reconstruction and axonal sprouting...
