Objective To characterize the acute and chronic behavioral, electrographic and histological changes of sustained seizures induced by kainic acid(KA ) ; and study the fuctions of Sema3C, Sema3 F in mossy fiber sprouting and synaptic reorganization in rats of KA -induced mesial temporal lobe epilepsy(MTLE) by detecting Sema3C, Sema3F mRNA expression. Methods Adult rats were made to be a model of synaptic reorganization TLE by intraventriculation of KA inducing status epilepticus. Behavioural evidence was obserned ; the electrophysiological was studied by electroencephalography(EEG); mossy fiber sprouting and pyramidal neuron loss and sprouting of granule cell were detected by Neo-Timm staining and Nissal staining. ; Sema3C/Sema3F mRNA expression was studied by in situ hybridization. Results Seizures started immediately after KA intraventriculatal injection and recurrid for up to one month. EEG showed a continual spikking limphy during SE and a singal, sharp waves , sharp-wave complexes and some times spikking limphy discharges during spontaneous recurrent seizures . Within the two weeks after KA-induced SE, cell loss in CA1 pyramidal cells and hilar neurons was prominent but limited. After two weeks of KA-induced SE, cell loss in these regions was markedly enhancing and defusing to CA2...
