Objective To investigate the genetic polymorphism of new five Y-STR loci DYS708, DYS712, DYS713, DYS714, DYS715 and two Y-STR loci of good polymorphism by demonstrated DYS576, DYS709 in Eastern Chinese Han population. and evaluate the forensic application of the seven loci . A multiplex PCR system is to be developed to amplify three Y-chromosomal STR loci DYS708,DYS713,DYS715.Methods Blood samples were collected from 108 unrelated Han ethnic group males living in Eastern China. A Chelex method was used for extracting genomic DNA. The PCR products were analyzed by non-denaturing polyacrylamide gel electrophoresis and visualized by silver staining.Certain allele of these seven Y-STR loci was sequenced and the allele ladders were constructed.Alleles of these seven Y-STR loci were nominated according to recommendations of the International Socity of Forensic Genetics.On account of above. a multiplex PCR system had been developed to amplify three Y- STR loci DYS708,DYS713,DYS715.Results In our study samples, 7, 13,7, 10, 7, 6 and 8 alleles were observed in the seven STR loci DYS708, DYS712, DYS713, DYS714,DYS715, DYS576, DYS709. The gene diversity (GD) of these loci were 0.7242, 0.9115, 0.8041, 0.8492, 0.8121, 0.7351 and 0.8202, respectively. The overall haplotype diversity reached 0.99999851. The sequencing results indicated that DYS708 contained two different repeat motifs, while the other loci had simple repeat motif. The core sequences were AGAT/AGCA, ATCT, TCTT, CTTTT, AAAG and CTTT respectively. A multiplex-PCR technique of typing three STR loci by silver saining had been constructed. It showed its typing result is right, well sensitive and stabe.Conclusion The results demonstrated these seven Y-STR loci showed high level of genetic polymorphisms and could be used in forensic science and human genetics study. The three Y-STR multiplex system would be very powerful for establishing Y-STR database, exploring human origin, paternity testing and personal identification.
