Xinnaoliantong capsule, consisted of Radix Salviae Miltiorrhizae, Herba Erigerontis, Rhixoma Polygoni Cuspidati, Crataegi Cuneatae, Folium Kaki, Radix et Rhizoma Acanthopanacis Senticosi and Radix Puerariae, is one of traditional Chinese medicine recipe for promoting blood flow and relieving pain. In the dissertation, method of quality control and the pharmacokinetics of puerarin in different decoction were studied.RP-HPLC methods were established to determine, simultaneously, puerarin, polydatin and scutellarin, salvianolic acid B and daidzein, cryptotanshinone and tanshinone II A, kaempferol in Xinnaoliantong capsule, respectively. The determination could be accomplished by different mobile phase system in Diamonsil C18 columns. The linear ranges were 23.20μg·mL-1 - 185.6μg·mL-1(r = 0.999 5), 15.20μg·mL-1 - 121.6μg·mL-1(r = 0.999 6), 4.600μg·mL-1 - 36.90μg·mL-1(r = 0.999 8), 19.20μg·mL-1- 153.5μg·mL-1(r = 0.999 5), 2.800μg·mL-1 - 22.40μg·mL-1(r = 0.999 7), 1.512μg·mL-1- 12.10μg·mL-1(r = 0.999 5), 0.918μg·mL-1 - 7.344μg·mL-1(r = 0.999 7), 1.700μg·mL-1 - 13.60μg·mL-1(r = 0.999 4), respectively. The recoveries were 100.0 %(RSD = 1.3 %), 98.7 %(RSD = 0.9 %), 100.4 %(RSD = 0.9 %), 100.8 %(RSD = 1.0 %), 99.2 %(RSD = 1.1 %), 100.3 %(RSD = 1.7 %), 100.4 %(RSD = 1.5 %), 100.0 %(RSD = 1.5 %), respectively. The content were 4.110 mg·capsule-1, 2.664 mg·capsule-1, 0.7580 mg·capsule-1, 3.899 mg·capsule-1, 0.5251 mg·capsule-1, 0.1530 mg·capsule-1, 0.0800 mg·capsule-1, 0.01656 mg·capsule-1, respectively.Determination of puerarin in rats plasma was applied to pharmacokinetic research by HPLC-UV. The HPLC separation was achieved on a Diamonsil C18 (200 mm×4.6 mm, 5μm, Dikma) column at 35℃. The mobile phase consisted of acetonitrile-0.05 % phosphonic acid solution (10.8:89.2, v/v) at a flow rate of 1.0 mL·min-1. The UV detection wavelength was set at 250 nm. The extraction recoveries exceeded 59.5 %. Linear calibration plot was obtained in the concentration range from 0.0800 to 12.00μg·mL-1(r≥0.99 88). Intra-day RSD and inter-day RSD were both less than 11.0 %. The method is simple, special, accurate and reproducible.Puerarin showed different pharmacokinetic characteristic in rats plasma after introgastric administration of different decoction, solution of decoction of Radix Puerariae and Xinnaoliantong capsule. Puerarin showed two-compartment model after introgastric administration of decoction of Radix Puerariae, and Xinnaoliantong capsule. After administration of decoction of Radix puerariae, Tmax was 0.528 h, t1/2β was 8.567 h, Cmax was 1.178μg·mL-1, AUC0-1 was 3.283μg·h·mL-1, AUC0-∞ was 3.985μg·h·mL-1. After administration of Xinnaoliantong capsule, Tmax was 0.639 h, t1/2β was 6.315 h, Cmax was 1.907μg·mL-1, AUC0-t was 3.926μg·h·mL-1, AUC0-∞ was 4.563μg·h·mL-1.
