Studies On The Active Fraction Of Danggui Shaoyao San

As a famous ancient prescription of traditional Chinese medicine,Danggui Shaoyao San (DSS)has been used for more than one thousand years.It is composed of Danggui(Angelica sinensis(Oliv.)Diels),Baizhu(Atractylodes macrocephala Koidz.),Baishao(Paeonia Lactiflora Pall.),Chuanxiong(Ligusticurn chuanxiong Hort.),FuLing(Poria cocos(Schw.)Wolf.), Zexie(Alisma plantago-aquatica L.var.orientale Sam.).It was mainly used for gynecological disease in ancient times,nowadays for senile gynecological disease,hypoimmunity and so on.This topic was based on the close cooperation of phytochemistry and pharmacology,to develop DSS into a new modern TCM medicine.After affirmed the pharmacodynamic action, DSS was separated into several parts which were screened on modal of pharmacology and the active part DSS-A-N-30 was selected as our target.A-N-30 was gained through optimizing the artwork of DSS-A-N-30.Seven compounds were isolated from A-N-30 by column chromatography methods using silica gel,Sephadex LH-20 and RP C-18.Six compounds were elucidated on the basis of chemical evidence and spectral analysis.Six compounds were identified as 4α- hydroxyl-albiflorin(2),paeoniflorin(3),2R(2α,4β,5α)-Spiro[2(3H)-furan-2,3' - 1H-pyrrolo[2,1 -c][1,4]oxazine]-5-carboxaldehyde,4,5-dihydro-4-hydroxy-5-hydroxylmethyl-(4), albiflorin(5),2R(2β,4α,5α)-Spiro[2(3H)-furan-2,3′-lH-pyrrolo[2,1-c][1,4]oxazine]-5-carboxal--dehyde, 4,5-dihydro-4-hydrox-y,5-hydroxylmethyl-(6),4″-hydroxyl-albiflorin(7).Among them, compounds 2,4,6 and 7 are four new compounds.
