Objective: To observe the effect of angelica shaoyao powder on renal tissue FN,LN,Col-Ⅳ in young mice with micropathological nephrotic syndrome,and to preliminarily reveal the mechanism of action of angelica shaoyao powder on kidneykidneysis at the molecular level,and to provide reliable experimental basis for its clinical application.Methods: 200 young male SD rats were divided into 5 groups according to random number table:(1)Normal group(2)Model group(3)Danggui Shaoyao San group(4)Candesartan group(5)Danggui Shaoyao Powder combined with Candesartan group(hereinafter referred to as combination group).40 in each group.Forty in each group.Except the normal group,all the other four groups were treated with doxorubicin by tail vein to construct the pathological model of renal micropathological changes.The modeling method was adopted for two times,that is,adriamycin was injected with 4mg/kg on the first day after the beginning of the experiment,and the baby mice were closely observed after the injection,and adriamycin was given again with 2mg/kg on the eighth day.From the 15 th day,each group was given the corresponding drug by continuous gavage for 42 d,and the experiment lasted for 8 weeks.Ten pups were randomly collected from each group on the 14 d,21d,42 d and 56 d of the experiment.Urine was collected for 24 h and the urine protein content was determined by BCA method for 24 h.After aortic blood was collected and kidney pathological sections were taken,various biochemical indicators were detected: plasma albumin(ALB),total cholesterol(TCH)and triglyceride(TG)levels were detected by automatic biochemical analyzer.ELASA method was used to detect the content of fibronectin(FN),laminin(LN)and col-iv collagen.The morphological structure of kidney tissue was observed by light microscopy.The changes of glomerular basement membrane and foot processes were observed by transmission electron microscopy.Summarize the data and analyze the results.Results: 1.At the 21 st day of experiment,the 24 h urinary protein content,FN,LN,Col-Ⅳ,TCH and TG in the model group were higher than those in the normal group,the ALB level was lower than that in the normal group,the difference was statistically significant(p < 0.05),under the electron microscope,the Glomerular basement membrane was uneven,thickened or exposed,the podocytes were deformed or fused,and there was noobvious increase of Mesangial Matrix.2.At the 42 nd day,the 24 h urinary protein content of Danggui Shaoyao San Group,candesartan group and combination group was lower than that of model group,and the ALB level was higher than that of model group(p < 0.05)The levels of TCH,TG,FN,LN,Col-Ⅳ in Danggui Shaoyao powder group and combination group were lower than those in model group(p < 0.05),and the recovery of biochemical indexes in combination group was better than that in single group(p < 0.05)The levels of FN,LN and Col-Ⅳ in model group were significantly higher than those in 21 days(p < 0.05).3.At the 56 th day,the biochemical indexes of each group were recovered continuously,and the biochemical indexes of the model group were deteriorated continuously(p < 0.05),and the indexes of FN,LN,Col-Ⅳ in Danggui Shaoyao powder group were close to the combination group,compared with Candesartan Group,the difference was statistically significant(p <0.05).Conclusion:1.Danggui Shaoyao Powder can reduce the urine protein content of NS pups by 24h;2.Danggui Shaoyao Powder can improve the level of ALB in young mice of NS;3.Danggui Shaoyao Powder can reduce the level of TCH and TG in NS pups;4.Danggui Shaoyang powder can significantly reduce the levels of FN,LN and COL-Ⅳ in NS pups.
