[Background and Objective] Cyclooxygenase (COX) is an important rate-limiting enzyme in the synthesis of prostaglandin (PGs) ,and there at least two isoenzymes have been found : COX-1 and COX-2. COX-1 is structure-based enzyme and express stably in most normal tissues, maintaining normal physiological function; COX-2 is an inducibaled enzyme,and rarely expresses in normal tissues, but it can express when the cells stimulated by various factors such as cytokines , growth factors or tumor promotor. COX-2 involves in inflammation or tumor physiological and pathological processes. This study is aim to investigate the relationship of COX-2 1759G>A (587Gly>Arg) gene with the susceptibility of lung cancer in Hubei Province population.[Method] A case-control study. The COX-2 1759G>A(587Gly>Arg) gene polymorphism of 200 lung cancer patients and 200 healthy people were detected using polymerase chain reaction - restriction fragment length polymorphism (polymerase chain reaction-restriction fragment length polymorphism, PCR-RFLP); The relationship of different genotypes with susceptibility of lung cancer was compared.[Results] The gene frequency distribution of COX-2 1759G>A(587Gly>Arg) polymorphism consistents with Hardy-Weinberg equilibrium, for the COX-2 1759G>A (587Gly>Arg)GA genotype, there was no statistically differences between lung cancer group and healthy control group (p = 0.092), and no statistically diferences in sex and age too, but for smoking, there is significant differences between the two groups(p=0.026,OR 为1.499,95%CI:1.125-3.231); COX-2 1759G>A gene polymorphism has no significant correlation with neither pathological type of lung cancer nor wheather lymph node metastasis or drinking.[conclusion] COX-2 1759G>A(587Gly>Arg) Polymorphism has a correlation with smoking volume ;COX-2 1759A allele maybe is a susceptible gene of lung cancer, and the exact molecular mechanism needs further study.
