Study Of The Correlation Among T-cell Subsets, Il-32, Il-6 And Hbv-infected Patients

Abstract/Summary:
Objective:To explore the changes of interleukin-32,interleukin-6 and T-cell subsets in patients with hepatitis B virus infection. The correlation among them and the clinical spectrum of the disease was analyzed. The results of serum IL-32 and IL-6 were also compared among groups based on the serum level of HBV viral load.Methods:Peripheral blood T-cell subsets were detected by flow cytometry (FCM); The serum levels of IL-32 and IL-6 were detected by enzyme linked immunosorbent assay (ELISA);liver function was assayed by automatic biochemistry analyzer; hepatitis B antigen/antibody semi-quantitative index was detected by Time-resolved fluorescence analyzer; HBV DNA was detected by Real-time fluorescent quantitative PCR.Results:1.IL-32 in different types of hepatitis B has significantly difference comparing with control group(F=108.494,P<0.001).IL-32 in acute hepatitis B is the greatest high than others. The level of IL-32 in serum was gradually increased in patients with mild, moderate and severe chronic hepatitis B.IL-6 in different types of hepatitis B has significantly difference comparing with control group(F=139.256,P<0.001),moreover the level of IL-6 in serum was gradually increased in patients with mild, moderate and severe chronic hepatitis B. The CD4+T and CD4+/CD8+T in different types of hepatitis B has significantly difference comparing with control group(F=3.07,P<0.05;F=4.00,P<0.05).2.The serum levels of IL-32 and IL-6 in the HBV-infected patients with HBV DNA positive group were higher than those observed in the control group, however, no significant difference was observed between them. The differences among various HBV viral load groups were also not statistically significant(P>0.05).3.The serum levels of IL-32 and IL-6 in various serum ALT groups have significantly difference comparing with control group(F=108.494,P<0.001).Among various serum ALT groups significant differences were observed(p<0.01).4.The serum level of IL-32 was negatively correlated with CD4+ T cell percentages and CD4+/CD8+T,the correlation coefficients r were separately-0.34 (P<0.05) and-0.60 (P<0.05).The serum level of IL-32 was positively correlated with the CD8+T cell percentages, the correlation coefficient r was 0.35 (P<0.05).The serum level of IL-32 was positively correlated with the ALT and TB, the correlation coefficients r were separately 0.50 (P<0.05) and 0.64 (P<0.05).The serum level of IL-32 was negatively correlated with Albumin,the correlation coefficient r was-0.38 (P<0.05).The serum level of IL-32 was no correlated with HBV viral load.5.The serum level of IL-6 was negatively correlated with CD4+T cell percentages and CD4+/CD8+T,the correlation coefficients r were separately-0.31 (P<0.05) and-0.57 (P<0.05).The serum level of IL-6 was correlated with the CD8+T cell percentages, the correlation coefficient r was 0.45 (P<0.05).The serum level of IL-6 was positively correlated with the ALT and TB, the correlation coefficients r were separately 0.31 (P<0.05) and 0.32 (P<0.05).The serum level of IL-6 was negatively correlated with Albumin, the correlation coefficient r was-0.40 (P<0.05).the serum level of IL-32 was no correlated with HBV viral load.6.The serum level of IL-32 was positively correlated with IL-6,the correlation coefficient r was 0.70 (P<0.05)Conclusion:1.The results indicate that the levels of IL-32 and IL-6 increases in HBV-infected patients. The levels of IL-32 and IL-6 increase with the aggravation of inflammation and synthetic function decrease in liver. Supposing that IL-32 and IL-6 play an important role in the HBV-infected patients.IL-32 and IL-6 are responsible for liver inflammatory injury process and diseases progression in HBV-infected patients.2.It seems that the changes of serum IL-32 and IL-6 level were not related to the HBV replication status.3.The serum level of IL-32 and IL-6 were negatively correlated with CD4+T cell percentages and CD4+/CD8+T,supposing that IL-32 and IL-6 maybe participate in the regulation of T-cell.
