Expression And Regulation Of Clara Cell 1o-kda Protein By Recombinant Murine Interleukin-10 In Allergic Rhinitis Of Murine Model

Objective: To investigate the expression of Clara cell 1O-KDa protein(CC10) and whether it is regulated by intranasal instillation recombinant interleukin-10(IL-10)in allergic rhinitis(AR)of murine mode1.Method: C57BL/6J murine were sensitized by intraperitoneal injection of ovalbumin (OVA)on days 0 and 5, then this was followed by daily challenge with intranasal instillation OVA from day 12 to day 19. the treatment groups were received intraperitoneal injection of dexamethasone(DXM)sodium phosphate or intranasal instillation recombinant interleukin-10(IL-10)15 minutes before instillation OVA, respectively. the control group was sensitized in the same way but challenged by saline instead of the OVA. The changes of eosinophilic cells, and goblet cells ,and expression of CC10 in nasal mucosa were examined by hematoxylin and eosin stain(HE),periodic acid-Schiff stain(PAS), and immunohistochemistry methods, respectively.Results: CC10 was mainly expressed by dome-like cells on the turbinate and ciliated epithelial cells on the septum, and decreased statistically in the OVA group compared with the control group, then came up markedly by the intraperitoneal injection of DXM or intranasal instillation recombinant IL-10, howerer, the effection of the former on CC10 was prior to the latter. eosinophilic cells and goblet cells were significantly increased while dome-like cells and ciliated epithelial cells were diminished in DXM and IL-10 group in contrast to control group, which were more statistically in OVA group.Conclusion:CC10,an endogenous factor, may take part in the development of allergic rhinitis, and it is up-regulated by exogenous IL-10 whose mechanism is not clear and needs further study.
