Objective:To investigate the therapeutical effect of scutellarin on hepatic fibrosis in rats. Methods:Immune hepatic fibrosis model of rats were made by intraperitoneal injecting porcine serum, twice a week for 13 weeks; On the ninth weekend, in addition to the normal control group, the rats were divided into liver fibrosis model group, scutellarin low dose group (25mg/kg), scutellarin middle dose group (50mg/kg), scutellarin high dose group (100mg/kg), colchicine group (positive control,0.15mg/kg); to feed drug from the ninth weekend, once a day for 28 days; All rats were sacrificed and were kept the blood and liver on the thirteenth weekend; divided the blood and fixed the liver; Serum levels of glutamic pyruvic transaminase, glutamic oxaloacetic transaminase, albumin, total protein, total bilirubin were detected by biochemical analysis; Serum levels of hyaluronic acid, laminin, Collagen TypeⅣwere detected by radioimmunoassay; Observed under light microscope hematoxylin-eosin liver biopsy and its pathological grade of hepatic fibrosis; Determined by immunohistochemistry of liver tissue typeⅠ,Ⅲcollagen content and calculated the color-rendering index. Result:Scutellarin can reduce the serum of glutamic pyruvic transaminase, glutamic oxaloacetic transaminase, total bilirubin, hyaluronic acid, laminin,Ⅳcollagen content, increase the serum of albumin, total protein content; Pathology observed by HE, the model of liver fibrosis was prepared successfully:Most structure of rat hepatic lobule was destroyed, there is extensive proliferation of fibrous tissue, hyperplasia fibrous tissue surrounded to form the varying sizes of false lobules and portal area expanded; Treatment groups repaired the organizational structure of the liver to different levels so that rats with liver fibrosis improved; Scutellarin can reduce liver tissue typeⅠ,Ⅲcollagen content. Conclusions:Scutellarin has therapeutical effect in hepatic fibrosis of rats.
