Object:platelet function and Concentration of plasma free calcium take part in many pathological changes, especially the functions and structures of heart, brain, and kidney. The study of them and how to regulate their abnormal function and concentration has become a hot topic in the domain of biological medicines .Recently, with the thorough study of the thorough pathogenesis and etiological factors of hypertension, we found that there exists abnormal changes of the function of platelet aggregation and the concentration of plasma free calcium. Then, we respectively observed the function of platelet aggregation and the concentration of plasma free calcium in SHR with left ventricular hypertrophy (LVH), after LVH, and congestive heart failure (CHF). Some studies showed that in SHR with LVH, the activity of protein kinase c increased significantly, and Scutellarin is a non-competitive PKC inhibitor, and it has multi-function that vessel dilation, increasement of blood flow, decreasement of peripheral vascular resistance, and anti- myocardial ischemia and anoxic. So, we used Scutellarin,Fosinopril and Enalapril to respectively intervene platelet aggregation and the concentration of plasma free calcium and myocardial remodeling in SHR, in order to discuss the relation between platelet aggregation, the concentration of plasma free calcium and myocardial remodeling, the mechanism of it, and the pharmacodynamics of regulation of the platelet function and improvement of myocardial remodeling by Scutellarin.Method: SHRs were bought in the animal center of cardio-angiopathy institute in FuWai Hospital of Chinese Medial Acadamy of Science. The SHR rats were divided randomly into NLVH group(1 month), LVH group(10 months) and heart failure group(18-20 months). And the normal WKYs were used in contrast of age pair-matching SHRs. BP-HR meter, weighing, transmission electron microscope, Fara-2/AM loading, fluorospectrophotometry and Born turbidimetry were all used to detect systolic blood pressure(SBP), heart rate(HR),hypertrophy index, ultrastructure of myocardial cells, concentration of platelet cytoplasm free calcium(PC[Ca2+]) and platelet aggregation(PAg). Meanwhile, 24 SHR rats were divided randomly into Scutellarin group, Fosinopril group, Enalapril group and control group. Then, the 8 weeks intervention treatment was carried on to observe the effect of PKCI and ACEI on SBP, HR, left ventricular weight/body weight(LVW/BW) , PC[Ca2+]i and PAg.Results:(1)ventricular hypertrophy hadn't occurred in 1 month SHR, but the character of apoptosis such as myocardial plasma concintration and karyopyknosis occurred in 1 month and 18-20 months SHRs. PC[Ca2+]i and PAg of 1 month SHRs increased a little than those of 1 month WKYs, but there was no significant difference between them(P>0.05).(2)There was significant differences between SBP of 10 months SHRs and 10 months WKYs(P<0.01), and they both had obvious character of left ventricular hypertrophy. LVW/BW of 10 months SHRs increased significantly that that of 10 months WKYs(P<0.01). It showed myocardial fiber hypertrophy and sarcoplasmic reticulum dilation , mitochondrion proliferation and changes of myocardial ultrastructures in 10 months and 18~20 months SHRs. And PC[Ca2+]i and PAg of the group had significant difference in contrast of pair-matching WKYs and 1 month SHRs(P <0.01)ï¼›Meanwhile, PC[Ca2+]i and PAg in 18-20 SHRs had increased significantly than those of 10 months SHRs(P<0.01).(3)Scutellarin, Fosinopril and Enalapril all could decreased significantly LVW/BW of SHRs with LVH, and improved the ultrastructure of myocardial hypertrophy. The 3 medicines could all decreased significantly PC[Ca2+]i and PAg of SHRs(P<0.01), especially in Scutellarin group. But Scutellarin had not obvious effect on SBP and HR of SHRs. Fosinopril and Enalapril could decreased significantly SBP of SHR, but they both had not obvious effect on HR.Conclusions:(1)Before obvious LVH, there was a obvious increasement in PC[Ca2+]i and PAg of SHRs; when de...
