Objective To study the correlation among H.pylori infection, the expression of matrix metalloproteinase-1 (MMP-1) and matrix metalloproteinase-10 (MMP-10) and gastric cancer. And to explore the effect of MMP-1 and MMP-10 induced by H.pylori infection in MGC-803 cells.Methods 1) ELISA were used to determine the IgG antibody of H.pylori. 2) The expression of MMP-1 and MMP-10 were detected by immunohistochemistry. And the correlation among H pylori infection, MMP-1 and MMP-10 expression and the clinical pathological features of gastric cancer were analysis by SPSS17.0 software. 3) The expression of MMP-1 and MMP-10 were detected by western blot after MGC-803 cell coculturing with H .pylori at different time.Result 1) The positive ratio of H.pylori infection was 77.5%(62//80)in gastric cancer which was higher than that in chronic gastritis with a ratio of 32.5%(13/40). And there was significant difference between them (p<0.05).2) MMP-1 expression was located in endochylema and cell membrane. The positive ratio of H pylori infection were 78.8%(63/80),95.0%(19/20) and 25.0%(10/40) in gastric cancer, lymph node metastasic gastric cancer and chronic gastritis, respectively. The expression of MMP-1 in lymph node metastasic gastric cancer was the highest ,,and in gastric cancer patients which was also higher than that in chronic gastritis. And there were significant meaning among metastasic gastric cancer , gastric caner and chronic gastritis (p<0.05).3) MMP-10 expression were located in endochylema and cell membrane. The positive ratio of H pylori infection were 83.4%(67/80),90.0%(18/20) and 30.0%(12/40) in gastric cancer, lymph node metastasic gastric cancer and chronic gastritis, respectively. The expression of MMP-10 in metastasic gastric cancer were higher than in gastric cancer. And in gastric cancer patients were also higher than in chronic gastritis (p<0.05). And there were significant meaning between lymph node metastasic gastric cancer and gastric caner, between gastric cancer and chronic gastritis (p<0.05).4) There were correlation among H.pylori infection and lymph node metastasis and clinic stage (p<0.05). And without correlation among H.pylori infection and age, gender, and differentiation degree; There were correlation among the expression of MMP-1 and MMP-10 and lymph node metastasis and clinic stage (p<0.05). And without correlation among the expression of MMP-1 and MMP-10 and age, gender, and differentiation degree.5) H.pylori infection was significantly correlated with the expression of MMP-1 and MMP-10 (r =0.525; r =0.412, P<0.05). MMP-1 was significantly correlated with the expression of MMP-10 (r =0.599, P <0.05).6) The exression of MMP-1 and MMP-10 were upregulated after induced by H .pylori compared to uninfected group (P <0.05). The expression of MMP-1 were upregulated in 12h after H .pylori infected and MMP-10 in 6h, respectively.Conclusions 1) The positive ratio of H.pylori infection in patients with metastasic gastric cancer was the highest, and in primarily gastric cancer it was higher than that in chronic gastritis. There were correlation among H.pylori infection and the metastasis and devolepment of gastric cancer.2) MMP-1 and MMP-10 played important roles during the metastasis and devolepment of gastric cancer. And there were correlation among H.pylori infection and the expression of MMP-1 and MMP-10.3) Upregulaiton of MMP-1 and MMP-10 caused by H.pylori infection in gastric cancer may be one of mechanism involving in the metastasis of gastric cancer.
