[Objective] The purpose of this study is to detect the expression of Nodal and Survivin, to analyzed their clinicalpatholical significance and to discuss their role with gastric cancer initiation and development[Materials and Methods] 50 cancer tissues were extracted from gastric cancer patients with complete follow-up information, and 20 normal tissue samples were extracted adjacent to these samples; The expression of Survivin and Nodal were detected through immunohistochemistry, the related clinical pathological data and follow-up information were collected. Data were analyzed by x2 test and and Spearman correlation analysis through SPSS version 13.0.[Results] In normal tissues, Nadal and Survin were expressed at low level,5% and 0% respectively. Nodal was highly expressed in 92.0% gastric cancer tissues (46/50), suggesting it was closely correlated with tumor differentiation, TNM staging and metastasis. The result of the univariate and multi variate analysis between the expression of the gene and postoperative metastasis suggested the expression of Nodal is an important prognostic index. The Survivin expression rate in gastric cancer 70.00%(35/50). In gastric cancer tissues, Survivin positivity rate was55.56%(10/18) in stagesⅠandⅡ, and 62.5%(10/16) in stage Ⅲand 93.75%(15/16) in stage IV respectively. There was statistical significance between them(x2=4.319,P<0.05). The expression rate of Survivin in high, middle and low histolog differentiation groups were 37.50%(3/8),72.41%(21/29), and 84.62%(11/13), respectively, there is statist significance between them(x2=4.875,P<0.05). We found Survivin positivity expression rate increased gradually with TNM stage upgrading and histological differentiation degrading. Survivin positivity rate was 71.05%(27/38) in patients with lymph node metastasis and 66.67%(8/12) in patients without lymph node metastasis. There was no significant association between Survivin expression rate and lymph node metastasis(x2=0.223,P>0.05). There was postive correlation between the expression Survivin and Nodal in gastric cancer(rs=0.452, P=0.002).[Conclusions]1. Expression of Nodal gene is a new and significant prognostic factor of patients with gastric cancer. High expression of this gene suggests lower of post operative metastasis and shortering of survival.2. Survivin is highly expressed gastric cancer tissues. No correlation was observed between Survivin positivity rate and lymph node metastasis, Survivin was closely correlated with differentiation,TNM staging and metastasis.3. The expression rate of Survivin and Nodal are postively correlated.
