ObjectiveThis project is based on theory of Traditional Chinese Medicine and clinical practice. Under the guidance of "xie zhuo jie du", we choose Rhubarb as study object which has good clinical curative effect and has been widely used, using metabonomics as technology support and RRLC-MS as analytical separation method, also combined with pharmacodynamics lab research, to explain the rationality and scientificity of Rhubarb in treatment of Chronic Renal Failure.MethodWe established rat animal model of Chronic Renal Failure, using Rhubarb Decoction to interfere, and detected related physiological, biochemical and pathological indicator to evaluate the effectiveness of Rhubarb in treatment of Chronic Renal Failure. We collected rat urine in different time point, applying RRLC-MS method to analyze the metabolite of rat urine sample, using statistical method such as PCA and PLS-DA to process data and acquire classified information and variables of differences in samples. Combining with the relation and substances origin of renal diseases, we screened out characteristic endogenous metabolite representing Chronic Renal Failure to study the mechanism of Rhubarb in treatment of Chronic Renal Failure.Results①Pharmacodynamic results shows:Rhubarb can reduce serum creatinine and urea nitrogen level, stabilize renal function, improve renal anemia symptoms, and correct electrolyte disorder of calcium, phosphorus, and magnesium etc. Rhubarb can also correct acidosis; ameliorate the disturbance of protein metabolism in the body; had some protective effect on renal compensatory hypertrophy and renal tubular injury and it is significant on delaying the development of Chronic Renal Disease.②Metabonomics results shows:Rhubarb can weaken or abolished the effect of the external stimulus (Adenine-induced chronic renal failure) in body, help normalizing the Metabolome of the animal in model. The mechanism of Rhubarb in treatment of Chronic Renal Failure. maybe relevant to potential biomarker such as Amino acid(Tyr,Trp,Phe,Leu), PA(18:0/18:1(9Z)), UDP-N-acetylmuramate, S-Methyl-5'-thioaden-osine etc.ConclusionThe effectiveness evaluation of metabonomics on Rhubarb in treatment of Chronic Renal Failure is consistent with the result of pharmacodynamic study, Metabonomics also reveals the potential biomarkers and metabolic pathways of the body which regulated by Rhubarb, enriches mechanism study on rhubarb in treatment of Chronic Renal Failure.
