Scorpio is dried whole of Buthus martensii karseh. It is mainly produced in Henan, Shandong, Hebei, etc. It smells micro-fishy and tastes salty. The one which is yellow, complete, body-ting, abdominal hard, new and bright is the best Scorpio.Scorpio has the effect of relieving convulsion and spasm, conteracting toxic substances and removing stasis and activating meridians to stop pain. And it is used to infantile convulsion, palsy and facial palsy, tetanic spasm, pyocutaneous disease, crewels, refractoriness headache and beriberoid disease.Recent studies have found that Scorpio, the tail of scorpion and scorpio toxin have effects on abdominal mass, ambury and they can not only destruct or inhibit the tumor cells but also can improve the body's own immune function. With the development of domestic and foreign bio-medicine, biochemistry and molecular biology, the scope of application of scorpio and the scorpio toxin is continually expanded and they have been used in clinical to cure a variety of difficult disease.Most of the modern researches of Scorpio venom are about its composition and pharmacological effects while there is still not a clear and unified understanding of the active ingredients of Scorpio. It is still need a further study on its pharmacological role and composition of protein, Amino, Fat-soluble chemical constituents and the relevance of various elements. This topic studies the fat-soluble components of the Scorpio and it can provide a reference of Scorpio's quality standards and GAP standards.The chemical constituents of the effective fraction were investigated intensively.6 compounds were isolated and purified by silica gel column chromatogram. On basis of their spectral data,5 of them were elucidated as pregn-5-ene-3,20-diol(I), cholesterol palmitote(Ⅱ), stearic acid(III), palmitic acid (Ⅳ), cholesterol(Ⅴ). Among them, compoundsⅠ,Ⅱ,Ⅲ,Ⅳ,Ⅴwere first isolated from Scorpio.Essential oil of Scorpio was studied by GC-MS. The substance in essential oil was identified by MS.47 substances were found, and among them 26 were identified. Most of them are fatty acids.Cholesterol was identified by TLC. Cholesterol spot was clear and it is not tailing and was well isolated from other constituents. Up to date, the quantification of cholesterol in Scorpion has not been found. Through this experiment we can find the high performance liquid chromatography method for the quantification of cholesterol in Scorpion. And this will provide theoretical foundation for pharmacology effect and determination quality standard of Scorpion.
