| Objective:We aim at observing effect of Chinese herbal medicine (CHM) compound Shenbaining on monocyte chemoattracctant protein-1 (MCP-1) in early stage diabetic nephropathy (DN) rat model, exploring mechanism of CHM compound Shenbaining on treating DN at early stage.Method:In this study, we used the left kidney removed and the STZ-induced DN rat model. Shenbaining extract was dissolved in saline, prepared into suspension and it was given 2.97g/kg, once/day,2ml/time orally to the CHM group. Lotensin was prepared into a suspension, and also was given 0.9gmg/kg,2ml/time,1 time/day, orally to the positive control group. The same volume of saline solution was given to the blank control group and the sham group in the same way. Then we observed the condition of their microalbumin in urine, kidney weight and body weight changes in rat model. Using immunohistochemistry and color image analysis system, the expression of MCP-1 was detected both qualitatively and quantitatively in the tissue of rats'kidney.Results:12 weeks later, it can be found in the blank control group that glomerular atrophy ball, balloon chamber slightly increased, the glomerular afferent artery with slight endothelial cell hyperplasia and fibrosis, basement membrane thickening slightly, distal convoluted tubule expansion, renal interstitial congestion, edema, blood cell infiltration and mild interstitial fibrosis. Compared with control group, the increased weight is higher in Chinese herb group significantly,while BMI of the kidney decreased in both groups,and it did not show significant deviation. After 12 weeks, Compared with the sham group and the blank control group,the microalbuminuria decreased in CHM group, and there were significant differences. Compared with the sham group, the expression of MCP-1 was significantly increased in the blank control group, CHM group and the positive control group,and there were significant differences. Compared with the blank control group,the positive expression in CHM group and the positive control group was much lower, and the difference was statistically significant. The expression of MCP-1 was of no significant difference between CHM group and positive control group. |
