Objective 1.Osteosarcoma cells were incubated with taxol (only one intervention point). Then the effect of taxol to the growth and apoptosis of osteosarcoma cells was observed. 2.Associating taxol with caffeine, observing the effect of association of them in the growth and apoptosis of osteosarcoma cells, and at last, investigating conceivable mechanism.Methods 1.Divide the concentration of taxol to 0.004ug/ml, 0.04ug/ml, 0.4ug/ml, 4ug/ml [ 1 ]. OS-732 cells were incubated with taxol at the above four concentration. Drawing cell proliferation curve. 2.Fixing the concentration of taxol to 1.2mg/ml (maximal clinical dosage). OS-732 cells were incubated with taxol at above concentration and Caffeine (2.0mmol/L, 5.0mmol/L). Drawing cell proliferation curve.Results 1.When the concentration of taxol reached 0.004ug/ml, OS-732 cells were inhibited obviously, which has obvious otherness compared to the control group (there is no intervention in this group). Moreover, the rate of apoptosis was more and more obvious as the concentration increased and time expanded. 2.Taxol (1.2mg/ml), combined with caffeine (5.0mmol/L), inhibited the growth of OS-732 cells more obviously than the group which was made up of taxol (1.2mg/ml) and caffeine (2.0mmol/L). Flow cytometry showed that, in groups which taxol and caffeine were combined, OS-732 cells stayed in G0/G1 phase, mitotic cycle was prolonged obviously, and the rate of apoptosis increased as time expanded.Conclusion 1.Although singly used, taxol inhibited osteosarcoma cells obviously. The inhibiting effect was enlarged as the consistence of taxol increased. The growth curvature appeared climbing. 2.The result of this study in vitro proved that caffeine, as a synergistic agent, combined with taxol, could inhibit the proliferation of OS-732 cells and accelerate its apoptosis. The mechanism of cooperation maybe that caffeine inhibits gene repair of osteosarcoma cells, and reverses the drug resistance of osteosarcoma cells to taxol. However, more clinical popularization and verification were needed to find out whether the association of the two drugs has similar effect in vivo.
