Objective Hydrogen peroxide-induced renal tubular epithelial cells (NRK52E) the establishment of oxidative damage model of Ginkgo biloba extract (EGB761) on rat renal tubular epithelial cells (NRK52E) Protective effects of oxidative damage, study EGB761 right renal epithelial cell apoptosis and to explore its mechanism.Methods In vitro cultured renal tubular epithelial cells in normal rats (NRK52E) is divided into a blank, hydrogen peroxide induced by different doses of EGB761 protection group, select 400μmol/L for 24 hours as a model to give different concentrations of EGB761 protection, detection SOD values to observe the cellular injury situation, the growth of cells was observed, streaming apoptosis rate detected, RT-PCR Detection of Survivin, Caspase-3 gene mRNA expression.Results (1)compared with the control group ,100-400μmol/L of hydrogen peroxide on renal tubular epithelial cells have significant injury, SOD was significantly lower (P<0.05), can induce apoptosis of renal tubular epithelial cells, MTT cell growth found in slow-flow AnnexinV-PI apoptosis rate increased (P <0.05). RT-PCR observation of Survivin gene mRNA expression did not change significantly (P<0.05), Caspase-3 gene mRNA upregulation (P <0.05). (2) and the control group and 400μmol/L hydrogen peroxide-induced group than from the cell growth curve and flow cytometry to different concentrations of EGB761 protection group was significantly inhibited hydrogen peroxide-induced apoptosis of renal tubular epithelial cells, RT -PCR observe the expression of Survivin gene mRNA increase (P<0.05), Caspase-3 gene mRNA expression reduced (P <0.05).Conclusion 1. Hydrogen peroxide with time and Concentration de pendent apoptosis induced by renal tubular epithelial cells; 2. EGB761 different concentrations of the cells significantly inhibited apoptosis; 3.EGB761 inhibit hydrogen peroxide-induced cell apoptosis Survivin may be through up-regulating genes, down regulation of Caspase-3 genes.
