Multiple Photonics Excitation Laser Scanning Confocal Microscope (MPELSCM) can provide more accurate medical basis about the early clinical diagnosis. Here, The collagen remodeling in photoaging which using intense pulsed light(IPL) treatment with different energy densities was observed in submicrometer to reveal the interaction of light with biological tissue effects. While because of the high-resolution and non-destructive diagnosis, Optical coherence tomography (OCT) can be used to measure the optical parameters and the structure of tissue. And it was used to observe the effect of IPL treatment and erbium:yttrium-aluminum-garnet (Er:YAG) laser treatment. The trend of the morphology and optical parameters were obtained.Firstly, an introduction to the diagnostic method of skin was briefly reviewed. The application and development of diagnosis based on Two-Photon microscopy (TPM)and OCT were reviewed, too. Further more, the purpose of our study was also presented. Based on the skin structure, the mechanism of photoaging and photorejuvenation were introduced. Then combining the instrument we used, the method of observation and estimation was discussed.Secondly, the photoaged skin was irradiated by an IPL and Er:YAG laser system in vivo under certain wavelengths with different energy densities. Meanwhile, the skin injury recovery process was observed by MPELSCM. By some image analysis methods, the morphological micro structure of collagen can be particularly achieved.Finally, The experiment research which major in interaction of IPL and Er:YAG laser in photoaged skin in vivo were made. And the skin injury recovery process was observed by OCT. The reasons concerning some complications were analyzed by the optical parameters'changes.
