ObjectiveTo study the effects of Dioscorea bulbifera rhizoma alone and compatibility with Angelica sinensis on grp78 and bad gene expressions of rat-livers. Discuss the mecha nism of liver injury caused by Dioscorea bulbifera rhizoma, which were reduced afte r compatibility with Angelica sinensis.MethodsThirty clean-level SD male rats were randomly (random number table method) di vided into three groups:used Dioscorea bulbifera rhizoma alone group (the Dioscore a bulbifera rhizoma group), Dioscorea bulbifera rhizoma combined with Angelica sine nsis group (compatibility group) and blank control group,10 in every group. Rats w ere given Dioscorea bulbifera rhizoma suspension (0.067g/mL) in the Dioscorea bulbi fera rhizoma group, while given Dioscorea bulbifera rhizoma compatibility Angelica s inensis suspension (including Dioscorea bulbifera rhizoma 0.067g/mL, Angelica sinens is 0.133g/mL) in compatibility group and 0.5% sodium carboxymethyl cellulose (CM C-Na) solution in blank control group,2mL, gavages,2times/d,14days. The grp78 a nd bad gene expressions of rat-livers were measured by quantitative real-time PCR (RTQ-PCR) after 14days with drug administration.ResultsAfter correction by gapdh, the grp78 and bad gene expressions in rat-livers of three groups were as follows:Dioscorea bulbifera rhizoma group grp78:2-ΔΔCt=339.9,bad: 2-ΔΔCt=256.2, compatibility group grp78:2-ΔΔCt=17.0,bad:2-ΔΔCt=9.9,blank control grou p grp78:2-ΔΔCt=1.0,bad:2-ΔΔCt=1.0. Compared to blank control group the grp78 and bad gene expressions of Dioscorea bulbifera rhizoma group were over-expressed(P<0. 01) and at the side of Dioscorea bulbifera rhizoma group the gene expressions of co mpatibility group were down-regulated(P<0.01). ConclusionsDioscorea bulbifera rhizoma up-regulated the grp78 and bad gene expression in r at-livers. After compatibility with Angelica sinensis, the enhancive effects of grp78 an d bad gene expressions were antagonized, which could be one of the mechanisms of liver toxicity were produced by Dioscorea bulbifera rhizoma but reduced after compa tibility with Angelica sinensis and protected the livers.
