This article uses the biology fermentation engineering method to research the strain separation, the purification, the selective breeding, the fermentation process and the chemical composition research for Pleurotus Spodotecus of medicinal fungus for the foundation of further pharmacology activeness and the clinical research.Applies the method of tissue isolation on the wild sporocarp of the Pleurotus Spodotecus to carry on the isolation and purification. We had breeded the vitality, metabolism ability exuberant and the fine strain of Pleurotus Spodotecus through carried on the strain breeding with the different nutritive elements and the culturing condition's inspection. In the liquid fermentation process research of Pleurotus Spodotecus , firstly we carried on the inspection to the liquid strain culture condition, obtained the best optimized culture medium. Through to technological conditions' inspection, we determined the best attire liquid volume, the table rotational speed, the raise temperature and the vaccination quantity. To simulate in the big industry production using the rocker strain culture to the ferment culture, through inspects the laboratory production fermentation culture termination index (PH of fermentation culture,hypha wet weight, dry weight, amino nitrogen content, protein content, polysaccharide content, ergosterol content, lovastatin content) to instruct the industrialization fermentation culture.By the silica gel column chromatography isolation techniques and the prepares the HPLC chromatograph technology separation to purify the chemical compositions in the Pleurotus Spodotecus, we obtained 3 monomers. The structures of the monomers were elucidated by the modern spectrodvopic analysis,such as ultraviolet, infrared, 1H NMR ,13C NMR and ESIMS.These monomers respectively were mannitol, ergosterol and lovastatin. And the lovastatin was discovered for the first time in the Pleurotus Spodotecus for the foundation of further expounding the active materials.The effective component ergosterol and lovastatin content in the Pleurotus Spodotecus carpophore were detected by HPLC. The results showed that the method was simple, accurate and could be used as quality control. |
