Theileriosis sergenti is a blood protozoal disease caused by Theileriidae, Theileria, Theileria sergenti. Recent years, Theileriosis result in economic losses seriously in animal husbandry with rising tendency,and paied the worldwide's attention. The study purpose was to establish the method of Dot-ELISA to detect antibody of Theileria sergenti, thereby to provide a more convenient and aeschynomenous and characteristic diagnostic method for clinical diagnosis of Theileriosis sergenti.The result shown optimal concentration of antigen was between 62.5μg/mL-15.625μg/mL;optimal concentration of serum dilution was 1:100;optimal condition of HRP-labeled sheep anti-bovine IgG was 1:2000;The blocking test, the crossing test and repeat test indicated that the method of Dot-ELISA was characteristic and steady; Detected 80 serum of cattle from hunchun by the Dot-ELISA and convention ELISA, the result indicated that the positive rate by Dot-ELISA was 17.9%, the positive rate by convention ELISA was 19.2%, the positive coincidence was 93.3%. negative coincidencewas 95.6%. and total coincidence was 98.7%.This experiment firstly established the Dot-ELISA by Chinese T.sergenti-p23 fusion protein. There were fortes not only high degree of specificity and high sensitivity of ELISA but also fewer antigen usage amount, quick and convenient about the Dot-ELISA, the diagnosis and prophylaxis of Theileriosis was provided dependable experiment method.
