Anemarrhena Bunge is a single species of single-plant in China, widely distributed in most parts of northern China,is one of commonly used traditional Chinese medicine,has high economic value of medicinal.In this study,first of all,through on-site investigation and expert advice,initially dividing the Zhimu type on the basis of their colour patterns and leaf-shaped variation,that is purple(or pink),white,wide leaf and narrow leaf,synthesized the result is purple-wide leaf,purple-narrow leaf and white-wide leaf,white-narrow leaf these four types.And then using RAPD and ITS molecular markers of different species of Zhimu form the source and type of molecular genetics research.In the study of RAPD,CTAB improved method was used to extract the total genome DNA of high quality of 16 Zhimu samples,after the application of RAPD analysis,by doing Polymorphism analysis research on different places anddifferent types Zhimu total genome DNi polymorphism analysis to search for Zhimu of RAPD molecular markers to determine the case within Zhimu genetic diversity.The results show that in the case Zhimu has rich DNA sequence polymorphism:in the group of different places,a total of 155 products were amplified by 26 arbitrary primers,of which 97 arepoly morphic products,5.96 polymorphic bands were amplifiedby each polymorphic primers on average,the average polymorphic rate is 62.58%;in different types Zhimu group,a total of 135 products were amplified by 26 arbitrary primers,of which 98 are polymorphic products,5.19 poly morphic bands were amplified by each polymorphic primers on average,the average poly morphic rate is 73.22%.Ntsys2.10 software using UPGMA cluster analysis: Zhimu from different origins can be divided into geographical distribution of two major groups,the first type including Shanxi Yuxian Zhimu and Hebei Pocang Zhimu,and Hebei Shexian Zhimu together as a sub-group,Heilongjiang Daqing Zhimu and Inner Mongolia Tongliao poly for another sub-group,in the second group in Shanxi Lingqiu and Hebei Neiqiu Zhimu as a sub-group together,and then Shanxi Dingbian Zhimu clustered together;in different types Zhimu group which comes from the same position in Hebei Pocang, firstly Zhimu by width divided into 2 groups and then the group was divided intowhite flowers group and purple flowers group,but in the wide leaves group classification obvious signs is not clear,this may be related to RAPD polymorphic bands not on the rich.In the study of ITS,still using the above DNA material,by doing ITS sequences research on Zhimu of different places and different types. Through reacting by PCR,about ?O0-800bp were amplified,and PCR products were sequenced by ABI310 sequencing machine.Anemarrhena asphodeloides Bunge' s Germplasm were analyzed by MEGA3.0:in the group of different places,Shanxi Dingbian Zhimu first differentiated from the Liliaceae, and then divide that in Shanxi Lingqiu Zhimu and Hebei Neiqiu Zhimu, further differentiation of the Inner Mongolia Tongliao Zbimu and Heilong jiang Daqing Zhimu,Finally,togethered with HebeiShexian,Shanxi Yuxian and Hebei Pocang Zhimu,a total of four branches of the type of reference: Patterns in different types of groups can be different based on flower colour will be divided into 2 major categories Zhimu,and further based on wide-width will be divided into Zhimu narrow leaves and Zhimu wide leaves,leaf morphological characteristics which are basically the same as the results of the survey. |
