| Among a set of monosomic alien addition lines of Beta corolliflora in sugar beet, a line, designated as M14, constituting of the normal 18 B. vulgaris chromosomes plus B.corolliflora chromosome 9, was found to have a high chromosome transmission frequency in successive generations. In order to sort and utilize the sugar beet M14 under free pollination, it was conducted to analyze the transmission frequency of 10 generations , investigate the diversity of the transmission frequency among plant lines or inflorescences in 5 generations and between the primary plant lines and those after 10 generations, and study the hereditary variation based on RAPD analysis. The main results were as below:.(1)The high transmission frequency of sugar beet M14 was heritable. But the transmission rate of the monosomic alien addition lines was different among plant lines or inflorescences. A few plants had the transmission ratio of no more than 50%. Most of the M14 lines in early generation can highly transfer. However, the transmission ratio decreased after several generations.(2)The PCR programme for RAPD suitable for M14 is optimized as follows: pre- pre-degeneration 94℃5min;degeneration 94℃1min, anneal 37℃1min,prolongation 72℃2min,40 cycles; prolongation 72℃7min. The reaction system of PCR included 10×PCR buffer 2μL , 100ng/μL template DNA 1μL ,2.5mmol/LdNTPs 1.6μL,25mmol/L MgCl2 2.0μL,20μmol/L primer 1.2μL,5U/μL Taq polymerase 0.25μL,ddH2O 11.95μL。20 primers were selected by the optimized programme.(3)The genotype of female plant and filial generation are not totally consistent. Also the genotype of different plants in one lines are variant. No asexuality line is produced by seed in the two M14 lines. |
