Isoenzyme and ISSR were applied successively to detect the genetic background of seven important aquacultural fishes, which are Mylopharyngodon piceus,Ctenph-aryngodon idellus, Hypophthalmichthys molitrix, Aristichthysnobilis, Megalobrama amlyceala, Erythroculter ilishaeformis and Siniperca chuatsi.The spots and enzyme belts of several isoenzymes - lactate dehydrogenase(LDH), esterase(EST),malate dehydrogenase(MDH), malic enzyme(ME), alcohol ehydrogenase (ADH), glutamate dehydrogenase (GDH) and peroxidase(POX) in seven fishes muscle organizations were analysed. Genetic distance and the similar coefficient of six kinds of Cyprinidae fish were calculated based on the number of spots and enzyme belts. The result of UPGMA was that Mylopharynodon piceus and Ctenophayngodon idellus clustered, Hypophthalmichthys molitrix and Aristichthys nobilis clustered, then these two groups formed a unit. Finally, this unit joined in another unit formed with Megalobrama amblycephala and Erythroculter ilishaeformis. The phylogenetic relationship was consistent with the result of taxonomy. This experiments showed that Mylopharyngodon piceus, Ctenpharyngodon idellus, Hypophthalmichthys molitr-ix, Aristichthys nobilis and Megalobrama amlycephala of seed farm maintains the genetic characteristics of natural stocks and are still good breed in Jiangsu. there are genetic differences among populations of Erythroculter ilishaeformis and Siniperca chuatsi because of reproductive isolation or geographical isolation.22 primers screnned from 100 primers could generate clear and reproductive bands. The repeat sequence of those primers are all based on di-nucleotide, which are (AG)n, (CT)n, (GA)n, (TC)n, (AC)n and (GT)n. The anchored sequences are at 3' end, but primer 884 was anchored at 5' end. The distribution of 22 primer to 7 fishes is different.
