| Shoot tissue culture now is considered of one of the best methods in micropropagation, which can assure plant genetic types. That's why some vegatables produced in industry by this way,and it is also more economic than other methods.Shoot tissue culture systems of six kinds of rosaceous ornamentals were builded,including Potentilla fruticosa " Red Robin" , Potentilla fruticosa "{Catherine Dykes", Cotoneaster horizontalis , Crataegus monogyna , Malus speclabilis"Hopa" , Prunus ceracifera "Nigra". A number of experiments were conducted to identify suitable procedure for in vitro shoot multiplication of these ornamentals. Different nutrient media(MS and WPM)and different plant hormones were studied in the experiment of different shoots' induction. Different concentration of different plant hormones,agar and sucrose were studied in initial culture , subculture and rooting. Most of explants,expect Cotoneaster horizontalis , axillary buds were able to sproute in low concentration cytokinin or no cytokinin .In subculture ,high concentration cytokinin , especially BA , always induced vitrification. So medium containing low cytokinin was suited.In rooting culture, IAA and IBA were better than NAA, however in these ornamentals which one is best can't be conclusion. This is due to diversity of improved variety.There are some problems affecting the application of forest trees tissue culture in industry. Vitrification is around in our research. Though sugar,agar and plant hormones were studied,these factors should not be key. For this , aerated micropropagation was studied in our research, which comes of photoautortophic micropropatation(sugar-free micropropagation). By ameliorating, it may provide the tissue cultural seedling micropropagation production possibility and reduce probability ofvitrification and contaminated. But lots of problems exerting effect on the cultural plant growing need further studying . |
