| The POD activity of wheat heads was evaluated after inoculated with spores of Fusarium graminearum. In the first six days, the POD activity of inoculated wheat heads was higher than non-inoculated heads during in resistant varieties; the POD activity also increased a little in medium-resistant variety; but had no difference compared with control in susceptible varieties. The protein profile of extracts was examined by FPLC. The results showed that one protein peak (designated P0) was bigger in resistant varieties When the sample were inoculated with spores or H202, but no difference in susceptible varieties, so protein P0 was related to wheat resisting Fusarium head blight. After non-inoculated samples were treated with H2O2, peaks of protein had the trend of left-moving in resistant varieties, but trend of right-moving in susceptible varieties. So, when H2O2 existed, enchancement of POD activity impelled low-weight substance to polymerize high-weight protein in resistant varieties, but no difference in susceptible varieties.Samples from callus of five wheat varieties with varying degrees of resistance against wheat head blight were examined by FPLC. The results showed that four absorbing peaks (designated P0, P1, P2and P3) got bigger after treated with Fusarium toxin (DON) in resistant varieties, but these absorbing peaks were smaller in susceptiblevarieties. Probably these proteins were related to wheat resisting Fusarium head blight. When the control samples of resistant varieties were treated with H202, their profile absorbing peaks was still similar to that of callus treated with toxin (DON). These also indicated the change of proteins was mediated by H2O2 and catalyzed by POD during wheat resistance against toxin, and no difference in susceptible varieties. The contents of salt soluble proteins had no obvious regularity in plants of 3 near-isogenic wheat lines with sumai-3 background after inoculation with spores of F.graminearum. When resistant line was inoculated, POD activity was higher than control, and POD activity was lower than the non-inoculated when medium-susceptibleand susceptible lines were inoculated in a long period. During the first eight days PO also increased when resistant line was inoculated with Fusarium, But no these phenomena in medium-susceptible and susceptible lines. |
