Chicken Anemia Virus(CAV) is the pathogen of ChickenInfectious Anemia(CIA). CAV belongs to Circoviridae. CAV genomeencodes three kinds of proteins: VP1(51.6KD), VP2(24.OKD) andVP3(13.6KD), respectively.VP1 is the only capsid protein of CAy, itcan induce chickens to produce immune protection with the help ofVP2 protein.A pair of primers was designed and synthesized according to thesequence of vp 1 gene. The fragment amplified by PCR technique wasligated into clone vectorsè†'UC18 and cloned in E.coli DH5- ct?Itwas sequenced via dideoxy-mediated chain-termination method.Sequence analysis and comparison with the vp 1 sequences reported inGeneBank Via DNA Blast showed that the identities are 95% or so.The differences resulted in the changes of amino acids encoded by vplgene. vpl gene was subcloned into pET3O(b~) vectors and induced toexpress in E.coli DE-3 by IPTG. Identification via SDS-PAGEshowed that vp 1 gene was expressed successfully and its molecularweight is about 51.6KD.The successful cloning and expressing of vp 1 gene become thebasis for the immunological studies of VP1 protein and themanufacture of genetic engineering vaccine of CAV.
