Chicken Anemia Virus(CAV ) is the pathogen of Chicken Infection Anemia(CIA). CAVbelongs to Circoviridae, and was first isolated by Yuasa of Japanese resercher in l979.CAV genome encodes three proteins:VP1(5l.6kD), VP2(24kD)and VP3(l3.6kD).VP3(Apoptin) can induce cellular apoptosis.We designed and synthesized two pairs of primers according to CAV genome sequence .Twofragments amplified by PCR technique were ligated into the complete CAV genome that wascloned into pUC18 vectors. Sequence analysis by dideoxy-mediated chain-termination methodshowed the cloned CAV had the similar sequence to the complete CAV genome in GeneBank.The cloning of the complete CAV genome is helpful to change its virulent protein gene,attenuate or eliminate virulence, and overcome the disadvantages of making vaccine withcommon methods. The study opens a new way of CAV attenuated vaccine reserches, and becomesa basis for CAV PCR diagnosis system and CAV genetic engineering vaccine.
